Transit amplification (TA) of progenitor cells maintains tissue homeostasis by balancing proliferation and differentiation. In testis, the germline proliferation is tightly regulated by factors present in both the germline and the neighbouring somatic cyst cells (SCCs). Although the exact mechanism is unclear, the epidermal growth factor receptor (EGFR) activation in SCCs has been reported to control spermatogonial divisions within a cyst, through downstream activations of Rac1-dependent pathways. Here, we report that somatic activation of the mitogen-activated protein kinase (Rolled/ERK) downstream of EGFR is required to synchronize the mitotic divisions and regulate the transition to meiosis. The process operates independently of the Bag-of-marble activity in the germline. Also, the integrity of the somatic cyst enclosure is inessential for this purpose. Together, these results suggest that synchronization of germ-cell divisions through somatic activation of distinct ERK-downstream targets independently regulates TA and subsequent differentiation of neighbouring germline cells.

Parkinson's disease (PD) represents a progressive neurodegenerative disorder with escalating global burden, with mechanistic studies revealing α-synuclein propagation through gut-brain axis, mitochondrial defects, and neuroinflammatory cascades driven by genetic-environmental interplay. Recent advancements in diagnostic paradigms have successfully combined α-synuclein seed amplification assays with multimodal neuroimaging techniques, achieving an impressive diagnostic accuracy of 92% during the prodromal stages of disease. Phase II trials highlight disease-modifying potential of α-synuclein-targeting immunotherapies (40% reduction in motor decline) and LRRK2 kinase inhibitors showing blood-brain barrier penetration.

Di-(2-ethylhexyl)-phthalate (DEHP) is a persistent environmental endocrine toxicant present in many products, and liver is the main target organ for DEHP metabolism. Long-term exposure to DEHP induces hepatic fibrosis, which is reversible in the early stages, while progresses to cirrhosis without timely intervention. Radiotherapy, a pivotal treatment for colorectal cancer, is compromised by tumor repopulation, which is characterized by accelerated growth and increased treatment resistance. Although radiation-induced DNA breaks eliminate most cells, a subset of polyploid giant cancer cells (PGCCs) evade death through massive genomic amplification, subsequently undergoing depolyploidization via a viral budding-like process to generate proliferative progeny. Critically, these PGCCs drive tumor repopulation and underpin therapeutic failure.

Short tandem repeat (STR) analysis is the cornerstone of forensic human identification due to its high discriminatory power and robustness. A concordance study was conducted to evaluate allele call consistency between two STR multiplex kits: PowerPlex® ESI 16 Fast (Promega Corporation, Madison, WI, USA) and Investigator®24plex GO! Kit (QIAGEN GmbH, Hilden, Germany). A total of 2000 buccal FTA cards (Whatman™ Flinders Technology Associates, Cytiva, Marlborough, MA, USA) samples previously typed with the PowerPlex® ESI 16 Fast Kit were re-analyzed using the Investigator® 24plex GO!
