Article Abstract

Embryonic stem cells can be incorporated into the developing embryo and its germ line, but, when cultured alone, their ability to generate embryonic structures is restricted. They can interact with trophoblast stem cells to generate structures that break symmetry and specify mesoderm, but their development is limited as the epithelial-mesenchymal transition of gastrulation cannot occur. Here, we describe a system that allows assembly of mouse embryonic, trophoblast and extra-embryonic endoderm stem cells into structures that acquire the embryo's architecture with all distinct embryonic and extra-embryonic compartments. Strikingly, such embryo-like structures develop to undertake the epithelial-mesenchymal transition, leading to mesoderm and then definitive endoderm specification. Spatial transcriptomic analyses demonstrate that these morphological transformations are underpinned by gene expression patterns characteristic of gastrulating embryos. This demonstrates the remarkable ability of three stem cell types to self-assemble in vitro into gastrulating embryo-like structures undertaking spatio-temporal events of the gastrulating mammalian embryo.

Recently, the research field revolving around the stem cell-based modeling of the human embryo gained particular momentum when the first integrated models were designed with the aim to recapitulate the development of the entire early human conceptus. The underlying driving force to reconstruct embryo-like structures is the prospect of a more comprehensive understanding of the fundamental processes controlling early human embryogenesis including their deregulation causing reproductive failures, and the endeavor to use these embryo models for drug testing and disease modeling. Although efforts will continue to create improved models with steadily increasing fidelity to human embryogenesis, the next phase focusing on the application of human embryo models as a platform to address particular scientific questions is currently being entered.

Pluripotent stem cells (PSCs) have been derived from various species, but most culture systems stabilize only a single PSC type. By contrast, epiblast cells in vivo exist along a continuum and interact dynamically with both embryonic and extraembryonic cells, interactions missing in standard PSC cultures. This absence limits the self-organizing potential of PSCs and leads to disorganized tissue formation in teratomas.
