A new metabolite, cyclic dipeptide, cis-(3S,8aS)-3-(3,4-dihydroxybenzyl)hexahydropyrrolo[1,2-a]pyrazine-1,4-dione, named JS-3 was isolated from Streptomyces sp. 8812 fermentation broth. Its chemical structure was established by means of spectroscopic analysis. A wide-range-screening study, which included inhibition assay of DD-carboxypeptidase/transpeptidase activity, determination of antibacterial, antifungal, and antiproliferative activities as well as free-radical scavenging was performed. To authors knowledge, this is the first isolation of such compound from natural sources and the first one from bacteria, Streptomyces.

DeoR family transcriptional regulators are widely distributed in Streptomyces genomes, but their precise functions and regulatory mechanisms in secondary metabolite biosynthesis remain poorly understood. In Streptomyces bingchenggensis, an industrial producer of milbemycins and nanchangmycin, we identified the DeoR-type regulator SbhR as a key regulator of the atypical two-component system AtcK/R. CRISPR interference-mediated inhibition of sbhR significantly decreased milbemycin production but enhanced nanchangmycin production.

Among the diversity of microorganisms, the rarest and least explored are microorganisms that live in conditions of high oxygen in the environment and can experience the effects of natural oxidative stress. Here we suggest that the actinobacteria of Lake Baikal, sampled in the littoral zone, may produce natural products with antioxidant activity. The current study aimed to assess the effects of experimentally increased amounts of oxygen and ozone on the morphology of actinobacteria, DNA mutations, and antioxidant potential.
