The mammalian orthoreovirus (reovirus) outer capsid undergoes a series of conformational changes prior to or during viral entry. These transitions are necessary for delivering the genome-containing core across host cell membranes. Reoviruses coordinate their replication and assembly through intricate spatial and temporal compartmentalization within host cells. In this study, we elucidate the dynamics of mammalian orthoreovirus (reovirus) core replication and viral particle assembly. Using high-resolution immunofluorescence confocal microscopy, we tracked input cores and de novo cores, revealing that input cores initially form peripheral, OC-negative factories that migrate inward while seeding independent peripheral factories.

Treeshrews, which share a closer common ancestor with primates and flying lemurs than with rodents, have been proposed as a better animal model for biomedical research compared to traditional rodent models. Zoonotic viruses like Mammalian orthoreovirus (MRV) pose potential public health risks, but their prevalence and characteristics in treeshrews remain underexplored. This study reports the first isolation of a novel MRV from common treeshrews (Tupaia glis) in Malaysia and investigates its biological properties.

Mammalian orthoreoviruses (MRVs) have a wide geographic distribution worldwide and have been detected from humans and a variety of animal species. This study represents the first isolation of MRV from sheep rectal swabs in China, with analyses of its molecular and pathogenicity characteristics. MRV-positive samples were inoculated into Madin-Darby bovine kidney (MDBK) cells, resulting in stable cytopathic effects (CPEs) after three generations of blind passage.
