Article Abstract

We describe a lentivirus-encoded chimeric receptor, termed extracellular vesicle (EV)-internalizing receptor (EVIR), which enables the selective uptake of cancer-cell-derived EVs by dendritic cells (DCs). The EVIR enhances DC presentation of EV-associated tumor antigens to CD8 T cells primarily through MHCI recycling and cross-dressing. EVIRs should facilitate exploring the mechanisms and implications of horizontal transfer of tumor antigens to antigen-presenting cells.

Immunoelectron Microscopy (IEM) is a technique that combines specific immunolabeling with high-resolution electron microscopic imaging to achieve precise spatial localization of biomolecules at the subcellular scale (< 10 nm) by using high-electron-density markers such as colloidal gold and quantum dots. As a core tool for analyzing the distribution of proteins, organelle interactions, and localization of disease pathology markers, it has irreplaceable value, especially in synapse research, pathogen-host interaction mechanism, and tumor microenvironment analysis. According to the differences in labeling sequence and sample processing, the IEM technology system can be divided into two categories: the first is pre-embedding labeling, which optimizes the labeling efficiency through the pre-exposure of antigenic epitopes and is especially suitable for the detection of low-abundance and sensitive antigens; the second is post-embedding labeling, which relies on the low-temperature resin embedding (e.

Background: The accuracy of CA19-9 detection results is very important for the early detection and treatment of pancreatic cancer, cholangiocarcinoma and other malignant tumors. At present, endogenous substances such as heterophilic antibodies will cause interference in the commonly used detection method is electrochemilumines-cence immunoassay, resulting in inaccurate results and affecting clinical diagnosis and treatment.

Methods: This paper reports a patient with an abnormal increase in serum CA19-9 level to explore how to accurately identify non-specific interference and anti-interference countermeasures.

Radiation therapy (RT) plays important roles in cancer treatment, and the efficacy of RT depends on the abscopal effect, which results in the regression of distant and untreated tumors through localized irradiation of a single tumor lesion. This effect is mediated by effector tumor antigen-specific T cells (ETASTs) activated by RT. Monitoring the radiation-induced changes in ETASTs can be used to predict the abscopal effect.

Nanoscale organization of integrin-mediated receptor crosstalk is crucial for controlling cellular signaling in cancer biology. Previously, interactions between integrin αvβ6 and receptor tyrosine kinases (RTKs) have been implicated in cancer progression, but the spatial regulatory mechanisms remain undefined. Here, we developed a programmable DNA origami-based platform for nanoscale control of heteroligand multivalency and spacing, enabling systematic investigation of αvβ6-RTK interactions in cancer biology.

Steroid-refractory gut acute graft-versus-host disease (SR-Gut-aGVHD) is the major cause of nonrelapse death after allogeneic hematopoietic cell transplantation. High numbers of donor-type IL-22+ T cells, IL-22-dependent dysbiosis, and loss of antiinflammatory CX3CR1hi mononuclear phagocytes (MNPs) play critical roles in SR-Gut-aGVHD pathogenesis. CEACAM1 on intestinal epithelial cells (IECs) is proposed to regulate bacterial translocation and subsequent immune responses in the intestine.
