Immunoglobulin A nephropathy (IgAN), the most prevalent primary glomerulonephritis globally, is characterized by mesangial IgA deposition and heterogeneous clinical trajectories. Historically, management relied on renin-angiotensin system inhibition and empirical immunosuppression, yet high lifetime kidney failure risk persists despite optimized care. This review synthesizes advances in molecular pathogenesis, highlighting how the traditional multi-hit hypothesis-while foundational for targeted therapy development-fails to capture IgAN's recurrent, self-amplifying nature.

Diffuse large B-cell lymphoma is the most common type of non-Hodgkin lymphoma (NHL) in humans, accounting for about 30-40% of NHL cases worldwide. Canine diffuse large B-cell lymphoma (cDLBCL) is the most common lymphoma subtype in dogs and demonstrates an aggressive biologic behaviour. For tissue biopsies, current confirmatory diagnostic approaches for enlarged lymph nodes rely on expert histopathological assessment, which is time-consuming and requires specialist expertise.

CD4 T follicular helper (T) cells support tailored B cell responses against multiple classes of pathogens. To reveal how diverse T phenotypes are established, we profiled mouse T cells in response to viral, helminth and bacterial infection. We identified a core T signature that is distinct from CD4 T follicular regulatory and effector cells and identified pathogen-specific transcriptional modules that shape T function.

We report 35 patients who had a leukemic phase of diffuse large B-cell lymphoma/high-grade B-cell lymphoma with MYC and BCL2 rearrangements, also known as double-hit lymphoma (DHL). There were 23 men and 12 women with a median age of 57 years (range, 29-82). Eight patients had an established DHL diagnosis and later developed a leukemic phase of disease and 27 presented with DHL and a leukemic phase of disease at initial diagnosis.

CAR-T cell therapy, as a representative technology in cancer immunotherapy, has demonstrated notable success in the treatment of hematologic malignancies; however, a significant proportion of patients fail to achieve sustained remission. Through the analysis of bone marrow sequencing data prior to CD19 CAR-T cell therapy, we identified cellular adhesion as a pivotal factor influencing clinical outcomes. We developed a model to predict B-ALL treatment efficacy based on the core genes associated with cellular adhesion, which was validated in our clinical cohort.
