Article Abstract

Neurons extend neurites with an increased synthesis of phosphatidylcholine (PC) that is not only a membrane component but also a functional regulator with specific fatty acid composition. To analyze the local synthesis of the PC molecular species within neurons, we combined a compartmentalized culture system with matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI-IMS). We observed that a newly synthesized PC, which contains exogenously administered palmitic acid-d, is accumulated at the cell bodies and the tips of the distal neurites. The local accumulation within distal neurites is formed by distinct metabolic activity from cell bodies, suggesting that the local extracellular composition of free fatty acid can be a key to regulate specific functions of each PC molecular species. We expect our simple method to be a starting point for more sophisticated in vitro analytical methods for unveiling detailed lipid metabolisms within neurons.

Study Question: Can testicular tissue from trans women (trans tissue) be used to create human testicular organoids?

Summary Answer: Testosterone-producing and cytotypic human testicular organoids with bicompartmental architecture can be successfully generated from trans tissue.

What Is Known Already: Testicular organoids are a promising tool for studying testicular function and the effects of toxicants. Immature testicular cells are currently the most efficient at forming organoids that closely recapitulate seminiferous tubule-like architecture and functions.

Background: The human intestinal epithelium is essential for nutrient absorption, barrier function, and immune system regulation. Although organoid cultures have advanced the field of intestinal biology, limitations regarding restricted access to the apical side remain, impeding drug administration and epithelial secretion analysis. To address these limitations, we developed an air-liquid interface (ALI) culture system using human induced pluripotent stem cells (iPSCs) to generate intestinal epithelium and evaluate epithelial polarity by analyzing extracellular vesicle (EV) secretion profiles.

Arbuscular mycorrhizal fungi colonization facilitates nitrogen uptake in cotton under nitrogen - reduction condition.

Background: Cotton is an economically important global crop, the yield and quality of which are strongly influenced by soil nitrogen. Low nitrogen use efficiency poses an important challenge to improve cotton yield and quality. The use of arbuscular mycorrhizal fungi (AMF) has been proposed as an effective solution to this challenge.

Bufadienolides, the primary active constituents of the traditional Chinese medicine Venenum bufonis, exhibit significant pharmacological activity. However, the potential pharmacokinetic effects of drug-drug interactions among these compounds under combination treatment have not yet been investigated. Thus, this study focused on three representative bufadienolides (resibufogenin, bufalin, and arenobufagin) as the research subjects and assessed the pharmacokinetic interactions among them under co-administration conditions.

Harnessing TGF-β signaling to improve testicular organoid development from dissociated testicular cells.

Background: Testicular organoids (TOs) have generated great interest in reproductive biology as a reliable experimental tissue model for pharmaco-toxicology studies and therapeutic applications. However, current TOs mostly fail to recapitulate the native testicular architecture and function, likely due to an imbalance between the spermatogonia and functional niche cells. TGF-β signaling pathway is a critical regulator of testis development that can be harnessed to regulate the testicular cells' behavior.
