Article Abstract

Background: Helicobacter pylori virulence is associated with different clinical outcomes. The existence of an intact dupA gene from tfs4b cluster has been suggested as a predictor for duodenal ulcer development. However, the role of tfs plasticity zone clusters in the development of ulcers remains unclear. We studied several H. pylori strains to characterize the gene arrangement of tfs3 and tfs4 clusters and their impact in the inflammatory response by infected gastric cells.

Methods: The genome of 14 H. pylori strains isolated from Western patients, pediatric (n=10) and adult (n=4), was fully sequenced using the Illumina platform MiSeq, in addition to eight pediatric strains previously sequenced. These strains were used to infect human gastric cells, and the secreted interleukin-8 (IL-8) was quantified by ELISA. The expression of virB2, dupA, virB8, virB10, and virB6 was assessed by quantitative PCR in adherent and nonadherent fractions of H. pylori during in vitro co-infection, at different pH values.

Results: We have found that cagA-positive H. pylori strains harboring a complete tfs plasticity zone cluster significantly induce increased production of IL-8 from gastric cells. We have also found that the region spanning from virB2 to virB10 genes constitutes an operon, whose expression is increased in the adherent fraction of bacteria during infection, as well as in both adherent and nonadherent fractions at acidic conditions.

Conclusions: A complete tfs plasticity zone cluster is a virulence factor that may be important for the colonization of H. pylori and to the development of severe outcomes of the infection with cagA-positive strains.

Neuropeptides (NPs) and their receptors (NPRs) play critical roles in modulating physiological processes and behaviors across species. While the transcriptional regulation of NP genes has been extensively studied, how NPRs contribute to context-dependent behavioral plasticity remains poorly understood. Here, we investigate the genomic features and expression patterns of NPRs in Drosophila melanogaster, leveraging comparative genomics, single-cell RNA sequencing (scRNA-seq), transcription factor (TF) network analysis, and empirical validation to uncover the regulatory mechanisms that involve NPRs and play roles in context-dependent adaptation.

Department of General Medicine, Chongqing University Central Hospital, Chongqing Emergency Medical Center, Chongqing Key Laboratory of Emergency Medicine, Chongqing, 400014, China.

Introduction: Acute Kidney Injury (AKI) is a clinical syndrome with rapid onset and poor prognosis, and existing diagnostic methods suffer from low sensitivity and delay. To achieve early identification and precise intervention, there is an urgent need to discover new precise biomarkers.

Methods: AKI samples were acquired from Gene Expression Omnibus (GEO) database.

Brown adipose tissue (BAT) is a key thermogenic organ that regulates energy expenditure through non-shivering thermogenesis. Rabbits (Oryctolagus cuniculus) are economically important livestock animals and popular model animals for medical research. However, the epigenetic mechanisms underlying BAT function remain poorly understood.

Background: The endoscopic transorbital approach (ETOA) has emerged as a valuable minimally invasive technique in skull base surgery, providing direct access to the anterior and middle cranial fossae. However, effective reconstruction of dural defects remains a significant technical challenge. This study evaluates the feasibility and anatomical characteristics of temporalis muscle flaps (TFs) and pericranial flaps (PFs) for dural reconstruction following ETOA, using cadaveric models.

The Mediterranean gorgonian octocorals are threatened by acidification, warming and marine heat waves. Phenotypic plasticity is critical for slow-growing gorgonians, as adaptation through natural selection might not be fast enough to cope with rapid environmental changes. DNA methylation (DNAm) is a type of (trans)generational phenotypic plasticity mechanism that may help slow-growing corals better withstand the effects of environmental changes by adjusting gene expression.
