A correct chromatin structure is important for cell viability and is tightly regulated by numerous factors. Human protein complex FACT (facilitates chromatin transcription) is an essential factor involved in chromatin transcription and cancer development. Here FACT-dependent changes in the structure of single nucleosomes were studied with single-particle Förster resonance energy transfer (spFRET) microscopy using nucleosomes labeled with a donor-acceptor pair of fluorophores, which were attached to the adjacent gyres of DNA near the contact between H2A-H2B dimers. Human FACT and its version without the C-terminal domain (CTD) and the high mobility group (HMG) domain of the structure-specific recognition protein 1 (SSRP1) subunit did not change the structure of the nucleosomes, while FACT without the acidic C-terminal domains of the suppressor of Ty 16 (Spt16) and the SSRP1 subunits caused nucleosome aggregation. Proteolytic removal of histone tails significantly disturbed the nucleosome structure, inducing partial unwrapping of nucleosomal DNA. Human FACT reduced DNA unwrapping and stabilized the structure of tailless nucleosomes. CTD and/or HMG domains of SSRP1 are required for this FACT activity. In contrast, previously it has been shown that yeast FACT unfolds (reorganizes) nucleosomes using the CTD domain of SSRP1-like Pol I-binding protein 3 subunit (Pob3). Thus, yeast and human FACT complexes likely utilize the same domains for nucleosome reorganization and stabilization, respectively, and these processes are mechanistically similar.

Patients with complex care needs present numerous challenges: the care they receive is often associated with more hospital admissions. The care provided to this group could benefit from being more goal-oriented and better integrated. However, strengthening a net-work of care for these patients starting from hospitalization remains a challenge.

This study investigates learning transfer processes in the teaching of pure tacts and intraverbals within the context of verbal behavior. The objectives were: to assess whether training pure tacts and intraverbals, through the inclusion of different stimuli, facilitates learning transfer to new impure tacts, and to determine whether one of these verbal operants (pure tact or intraverbal) better promotes learning transfer. The sample included 54 children aged 11-12 years, using a within-subjects experimental design with pre-post measures.

Short Tandem Repeats (STRs) are the standard technique used in forensic genetics for individual identification due to their high polymorphism and robustness. Although Capillary Electrophoresis (CE) enables the analysis of many STRs, Next-Generation Sequencing (NGS) offers enhanced resolution and the ability to detect STRs' isoalleles and their flanking regions, enhancing the discrimination power of this analysis. Despite the fact that STR kits for NGS are well standardized for evaluating forensic samples, there is no data on their effectiveness in differentiating monozygotic (MZ) twins, which are indistinguishable by CE.

Histone methylation (HMT), the enzymatic addition of methyl groups to specific histone residues by histone methyltransferases, constitutes a key regulatory mechanism in gene expression and post-translational modulation. Although studies have explored HMT's role in oncogenesis and other organ-specific disorders, HMT is now implicated in the pathogenesis of kidney diseases. A broad spectrum of experimental models, including both and systems, has demonstrated the involvement of HMT alterations in diverse renal pathologies such as acute kidney injury, renal fibrosis, diabetic nephropathy, lupus nephritis, polycystic kidney disease, kidney stones, renal cell carcinoma, and immunoglobulin A nephropathy.
