Article Abstract

A chemically cross-linked but remarkably (re)processable shape-memory polymer (SMP) is designed by cross-linking poly(ε-caprolactone) (PCL) stars via the efficient triazolinedione click chemistry, based on the very fast and reversible Alder-ene reaction of 1,2,4-triazoline-3,5-dione (TAD) with indole compounds. Typically, a six-arm star-shaped PCL functionalized by indole moieties at the chain ends is melt-blended with a bisfunctional TAD, directly resulting in a cross-linked PCL-based SMP without the need of post-curing treatment. As demonstrated by the stress relaxation measurement, the labile character of the TAD-indole adducts under stress allows for the solid-state plasticity reprocessing of the permanent shape at will by compression molding of the raw cross-linked material, while keeping excellent shape-memory properties.

Background: Thoracic aortic dissection (TAD) is a fatal cardiovascular disease characterized by vascular smooth muscle cell (VSMC) phenotypic remodeling and inflammation. Cannabinoid 1 (CB1/Cnr1) receptor has been implicated in cardiovascular diseases, but its role in TAD and its downstream signaling remains unclear. Genistein (Gen), an isoflavone abundant in legumes, has demonstrated promising therapeutic potential in cardiovascular diseases such as vascular calcification, hypertension, and atherosclerosis due to its anti-inflammatory, antioxidant, and metabolic regulatory properties.

Triazolinedione (TAD) derivatives have been commonly utilized as protection and labeling reagents for indole and phenol moieties a reversible ene-type reaction. Previous studies showed that the TAD probes could selectively modify tyrosine and tryptophan side-chains within proteins and peptides under distinct pH conditions. Here, we report a pH-controlled regioselective rapid ene-type reaction (RRER) between TAD and 5-hydroxyindole, where the modification occurs on the C4 position rather than the C3 of inactivated indole rings.

Obesity is a significant risk factor for thoracic aortic dissection (TAD), as supported by UK Biobank data showing obese individuals have a higher risk of TAD. The study investigates leptin, a hormone elevated in obesity, and finds that hyperleptinemia is common in TAD patients, suggesting its role in disease pathogenesis. Using leptin-knockout mice, it is demonstrated that exogenous leptin exacerbates TAD by promoting phenotypic transitions in vascular smooth muscle cells (VSMCs).

The combination therapy of mirabegron (MIR) and tadalafil (TAD) has gained significant attention in clinical practice for the treatment of Benign Prostatic Hyperplasia as well as overactive bladder syndrome. This study demonstrates the development and validation of two analytical methods for the concurrent determination of tadalafil and mirabegron in their synergistic combo therapy. Spectrophotometry and stability-indicating HPLC were employed for the accurate quantification of both drugs.
