During epileptic seizures, brain activity and connectivity undergo dramatic changes. Brain networks transition from a balanced resting state to a hyperactive and hypersynchronous state. However, the mechanisms driving these state transitions remain unclear.

Background: Evidence indicates that the interplay between pain and anxiety poses clinical challenges for the evaluation and management of chronic pain, yet effective therapies for these comorbidities are limited. This study aimed to elucidate the effects and mechanisms of electroacupuncture (EA) on pain-anxiety comorbidities.
Methods: Mice injected with Complete Freund's adjuvant (CFA) in the ipsilateral hind paw developed persistent inflammatory pain and anxiety-like behaviors, as assessed by the von Frey, open field, elevated plus maze, and novelty-suppressed feeding tests.

The TWIK-related K channel (TREK-1), a member of the two-pore domain potassium(K2P) family, is characterized as a "leaky potassium channel" and is integral to the maintenance of the resting membrane potential. As the most abundant cell type in the central nervous system, astrocytes play important roles in the development of epilepsy by regulating the release of glutamate and the function of potassium channels. Previous studies have revealed that TREK-1 is involved in a range of neurological diseases, including epilepsy.

Acrylamide (ACR) is a pervasive environmental and workplace contaminant with established neurotoxic effects but unclear pathogenic mechanisms. In this study, we screened for potential ACR binding targets associated with neurotoxicity and identified the astrocytic glutamate transporter EAAT2. Molecular docking and dynamics simulations revealed that ACR interacts stably with the glutamate-binding pocket of EAAT2, potentially impairing transport function.
