Article Abstract

Background: Adipose tissue damage of cryopreserved fat after autologous fat transfer is inevitable in several processes of re-transplantation. This study aims to compare and analyze the survivability of adipocytes after thawing fat cryopreserved at -20℃ by using thawing methods used in clinics.

Methods: The survival rates of adipocytes in the following thawing groups were measured: natural thawing at 25℃ for 15 minutes; natural thawing at 25℃ for 5 minutes, followed by rapid thawing at 37℃ in a water bath for 5 minutes; and rapid thawing at 37℃ for 10 minutes in a water bath. The survival rates of adipocytes were assessed by measuring the volume of the fat layer in the top layers separated after centrifugation, counting the number of live adipocytes after staining with trypan blue, and measuring the activity of mitochondria in the adipocytes.

Results: In the group with rapid thawing for 10 minutes in a water bath, it was observed that the cell count of live adipocytes and the activity of the adipocyte mitochondria were significantly higher than in the other two groups (P<0.05). The volume of the fat layer separated by centrifugation was also measured to be higher, which was, however, not statistically significant.

Conclusions: It was shown that the survival rate of adipocytes was higher when the frozen fat tissue was thawed rapidly at 37℃. It can thus be concluded that if fats thawed with this method are re-transplanted, the survival rate of cryopreserved fats in transplantation will be improved, and thus, the effect of autologous fat transfer will increase.

Towards blood on demand: Rapid post-thaw isolation of red blood cells from multicomponent cryoprotectants.

Severe blood loss due to trauma, anaemia, or chemotherapy necessitates immediate red blood cell (RBC) transfusions. The short shelf-life of RBCs at 4 °C complicates emergency supply management. Glycerol is the state-of-the-art cryopreservative for RBCs but the time from thawing to transfusion is more than 1 hour, due to the slow, extensive washing process.

Background: Natural killer (NK) cell immunotherapy is a promising approach for cancer treatment. However, its extensive clinical application was limited to the large-scale clinical-grade expansion of NK cells. In this study, we expanded NK cells from healthy donor's peripheral blood mononuclear cells (PBMCs) using a newly designed K562 feeder cell line.

Background: Adenomyosis is characterized by ectopic endometrial tissue, which causes dysmenorrhea, menstruation, and infertility, significantly impairing quality of life. Current treatments-including medications, surgery, and interventional methods-all have limitations. Cryoablation, a technique that induces tissue necrosis through ultra-low temperature exposure followed by rapid thawing, has demonstrated efficacy in treating solid tumors (e.

Thermo-responsive hydrogels can dynamically modulate incident light, providing a broad prospect for development of smart windows, which are of pivotal importance for energy conservation in buildings. However, these hydrogels normally exhibit slow response speed and tend to contract over extended phase transition, compromising structural integrity of smart windows. In this study, a solid-liquid switchable thermochromic hydrogel, denoted as SL-PNIPAm, was synthesized by cross-linking PNIPAm with AMEO through dynamic imine bonds.

Conventional polyvinyl alcohol (PVA) hydrogels with isotropic polymer networks usually lack selective responses to external stimuli, which significantly limits their applications in smart devices. In this work, a series of double network (DN) composite PVA hydrogels reinforced with cellulose nanocrystal (CNC) were prepared by in situ photopolymerization of functional monomers and freeze-thawing (F-T). The incorporation of CNC and the DN structure resulted in the high tensile strength of the hydrogel (1.
