The intestinal epithelium forms a barrier protecting the organism from microbes and other proinflammatory stimuli. The integrity of this barrier and the proper response to infection requires precise regulation of powerful immune homing signals such as tumor necrosis factor (TNF). Dysregulation of TNF leads to inflammatory bowel diseases (IBD), but the mechanism controlling the expression of this potent cytokine and the events that trigger the onset of chronic inflammation are unknown. Here, we show that loss of function of the epigenetic regulator ubiquitin-like protein containing PHD and RING finger domains 1 (uhrf1) in zebrafish leads to a reduction in tnfa promoter methylation and the induction of tnfa expression in intestinal epithelial cells (IECs). The increase in IEC tnfa levels is microbe-dependent and results in IEC shedding and apoptosis, immune cell recruitment, and barrier dysfunction, consistent with chronic inflammation. Importantly, tnfa knockdown in uhrf1 mutants restores IEC morphology, reduces cell shedding, and improves barrier function. We propose that loss of epigenetic repression and TNF induction in the intestinal epithelium can lead to IBD onset.

Background: The COVID-19 pandemic disrupted the provision of sexual and reproductive health services, including contraceptive and family planning (FP) services. The World Health Organization conducted a multi-country study in India, Nigeria and Tanzania to assess the impact of the pandemic on the health system's capacity to provide contraceptive and FP services. In this paper, we share the results of a qualitative study aimed at understanding clients' perspectives at the primary healthcare level on accessing contraceptive services in COVID-19-affected areas in the three aforementioned countries.

Entomopathogenic fungi can precisely inhibit the cellular and humoral immune responses of host insects by secreting effector proteins, allowing them to overcome the innate immune barriers of their hosts. Nodule formation is an immune response primarily mediated by insect hemocytes, which can rapidly and efficiently capture invading pathogenic fungi in the hemocoel. However, the molecular mechanisms by which fungi inhibit insect nodule formation through the secretion of effector proteins remain unclear.

Epilepsy is a neurological disorder that shows strong genetic control on the timing and onset of symptoms and drug response variability. Some epilepsy syndromes have clear monogenic mutations but genes with control on the phenotype and severity of the disorder and drug sensitivity are present in the whole genetic profile. Genetic modifiers are not the cause of epilepsy but control significant networks such as synaptic plasticity and ion channels and neurodevelopment and neuroinflammation and therefore the reason why two individuals with the same primary mutations have different clinical courses.

Ulcerative colitis (UC), a chronic inflammatory bowel disease (IBD), is characterized by disruption of intestinal barrier function and complex inflammatory manifestations locally and systemically. Although anti-tumor necrosis factor-α (TNF-α) agents such as Infliximab (IFX) are effective in treating IBD, their intestinal tissue concentration has been regarded as determinant of therapeutic efficacy while was restrained by the large molecular weight. Considering the enhanced expression of human neonatal Fc receptor (hFcRn) in UC tissues, we attempted to deliver the therapeutic entity of IFX into UC tissues by developing a novel dual-acting IFX Fab-F8 (IFX-F8) fusion protein for UC treatment.
