Article Abstract

Seven new prenylated phenols, five novel phenols (1-5) with polycyclic skeleton and two new phenols (6 and 7) with a carbon chain, along with one known compound (8) were isolated from the fruiting bodies of Ganoderma cochlear. The structures of new compounds were elucidated by the spectroscopic technologies, X-ray crystallography analysis and chiral HPLC chromatography. All compounds showed antioxidant effect in radical scavenging assays and a plausible biosynthetic pathway for 1-8 was proposed.

Oncogenic KRAS drives ~30% of solid tumours, yet the only approved G12C-specific drugs benefit ≈ 13% of KRAS-mutant patients, leaving a major clinical gap. We sought mutation-agnostic natural ligands from Ziziphus lotus, whose stereochemically rich phenolics may overcome this limitation by occupying the SI/II (Switch I/Switch II) groove and locking KRAS in its inactive state. Phytochemical mining yielded five recurrent phenolics, such as (+)-catechin, hyperin, astragalin, eriodictyol, and the prenylated benzoate amorfrutin A, benchmarked against the covalent inhibitor sotorasib.

Two previously unreported anthraquinones, named sintenisiquinones A (1) and B (2), were isolated from the MeOH extract of Asperula sintenisii together with five iridoid glycosides, five flavonoids, as well as three phenolic acid derivatives. Their chemical structures were elucidated on the basis of extensive 1D (H and C) and 2D (COSY, HSQC, and HMBC) NMR spectroscopy and HRESIMS. Compounds 1 and 2 represent rare type of anthraquinone derivatives bearing a prenyl group cyclized to dihydrofuran ring.

Prenylated phenolics are plant-derived compounds with antimicrobial activity against methicillin-resistant (MRSA), acting by targeting membranes resulting in fast permeabilization. Studies quantifying their membrane permeabilization capacity are lacking, limiting our understanding of the structural properties driving this effect. This study evaluated antimicrobial activity and permeabilization efficacy of 36 C- and O-prenylated phenolics, including 11 C- and O-prenylated phenolics chemically synthesized for this study.

(jackfruit) is widely distributed in subtropical and tropical regions, and some phytochemicals isolated from this species have demonstrated anti-proliferative effects. However, its impact on triple-negative breast cancer (TNBC) and HPV-related cervical cancer models remains unclear. This study evaluated the phytochemical profile and anticancer activity of an ethanolic extract from leaves (AHEE) in the TNBC cell line MDA-MB-231 and in the HPV-16 murine cancer cell line TC-1.
