Most human polymorphisms are neutral or slightly deleterious, but some genetic variation is advantageous and maintained in populations by balancing selection. Considered a rarity and overlooked for years, balanced polymorphisms have recently received renewed attention with several lines of evidence showing their relevance in human evolution. From theoretical work on its role in adaptation to empirical studies that identify its targets, recent developments have showed that balancing selection is more prevalent than previously thought. Here we review these developments and discuss their implications in our understanding of the influence of balancing selection in human evolution. We also review existing evidence on the biological functions that benefit most from advantageous diversity, and the functional consequences of these variants. Overall, we argue that balancing selection must be considered an important selective force in human evolution.

Background: The development of specific fluorescent probes for cancer cell discrimination holds significant promise for advancing cancer diagnostics. Gut barrier loss exacerbated gut microbiota dysbiosis by permitting pathogenic blooms, while gut microbiota dysbiosis caused the development of gut mucosal wounds by reducing mucus and breaking down epithelial tight junction. Current therapies combating colitis often fail to address both gut barrier dysfunction and microbial imbalance. Herein, inspired by natural gut mucus, a dual-crosslinked hydrogel (HSMP-LA) composed of thiol/maleimide-modified hyaluronic acid together with co-loading of antimicrobial ε-polylysine (ε-PL) and larazotide acetate (LA) had been developed as an injectable artificial gut mucus to simultaneously restore barrier integrity and modulate gut microbiota.

Acute graft-versus-host disease (GVHDa) is one of the leading causes of morbidity and mortality after allogeneic hematopoietic stem cell transplant (HSCT) patients. While the first-line consensus treatment has been based on systemic corticosteroid therapy for many years, ruxolitinib has recently been approved and has become the standard second-line treatment. Nevertheless, the effectiveness of ruxolitinib remains limited to 40 % of cortico-resistant patients, raising the crucial question of selecting a third-line treatment.
