Imidazopyridine 1 was identified from a phenotypic screen against P. falciparum (Pf) blood stages and subsequently optimized for activity on liver-stage schizonts of the rodent parasite P. yoelii (Py) as well as hypnozoites of the simian parasite P. cynomolgi (Pc). We applied these various assays to the cell-based lead optimization of the imidazopyrazines, exemplified by 3 (KAI407), and show that optimized compounds within the series with improved pharmacokinetic properties achieve causal prophylactic activity in vivo and may have the potential to target the dormant stages of P. vivax malaria.

Incorporating bioaccessibility into health risk assessments enhances the accuracy of exposure estimates for heavy metal (HM) pollution, supports targeted remediation, and informs public health and policy decisions, particularly for vulnerable populations. Because HM bioaccessibility depends on local soil and geographic characteristics, identifying its relationship with soil properties is crucial for assessing soil pollution potential. Although HM concentrations can be measured relatively easily, bioaccessibility requires complex laboratory procedures, limiting routine applications in regulatory contexts.

Importance: Advances in diagnostics have enabled the detection of more gastrointestinal pathogens, but misuse of diagnostics can lead to inappropriate antibiotic use and excess financial burdens. Ensuring appropriate use of diagnostics is crucial for optimizing patient care and promoting stewardship of health care resources.
Objective: To elicit parents' and clinicians' perspectives on expectations for care of pediatric diarrhea with a focus on diagnostic testing and to evaluate the potential for an electronic clinical decision support tool (ECDST) to improve appropriate use of diagnostics.

Donor-acceptor-donor (D-A-D) thiophene-based compounds, characterized by thiophene as a donor unit and benzothiadiazole (Bz) as an acceptor, represent an emerging class of theranostic agents for imaging and photodynamic therapy. Here, we expand this class of molecules by strategically varying the position of the electron-accepting unit within the oligothiophene (OT) backbone structure, realizing a series of different push-pull architectures (A-D, D-A-D, and D-A). This rational design allows for precise modulation of key photophysical parameters, including absorption and emission spectra, molar absorption coefficient, charge separation, and frontier molecular orbitals.
