Amyloid-β (Aβ) is implicated in the pathophysiology of Alzheimer's disease (AD) and plays a significant role in neuronal degeneration. Aβ in solution is essential during the initial stages of developing lead compounds that influence Aβ fibrillation. The tendency of the Aβ peptide to misfold in solution is correlated with the etiology of AD.

Introduction: The role of triglycerides in Alzheimer's disease dementia (ADD) progression remains unclear. This study aimed to investigate how triglyceride levels influence the relationship between amyloid-beta (Aβ) deposition, hippocampal atrophy, and cognitive decline in individuals with mild cognitive impairment (MCI) and early-stage ADD.
Methods: A total 188 older adults (170 with MCI, 18 with early ADD) from the Gwangju Alzheimer's Disease and Related cohort underwent amyloid PET and structural magnetic resonance imaging.

The diagnostic performance of the Amyloid Probability Score 2 (APS2; the algorithmic result of the PrecivityAD2™ blood test) was originally trained and validated in two cohorts of cognitively impaired (CI) individuals and is in clinical use. Here, we further test the repeatability and reliability of the prespecified APS2 algorithm and cut point in an independent cohort. APS2 diagnostic performance was determined using amyloid positron emission tomography (PET) as the reference standard.

Aggregation of the amyloid-β peptide (Aβ) characterises and probably causes Alzheimer's disease. While lipid-mediated Aβ aggregation has been extensively studied for the 40-residue variant Aβ40, the interaction of the 42-residue variant Aβ42 with membranes has received less attention. Our time-resolved infrared spectra demonstrate that Aβ42 oligomers preserve their β-sheet structure in aqueous solution also in a membrane-mimicking environment consisting of either 1-hexadecanoyl-2-(9Z-octadecenoyl)-sn-glycero-3-phosphocholine (POPC, zwitterionic) or 1-hexadecanoyl-2-(9Z-octadecenoyl)-sn-glycero-3-phospho-(1'racglycerol) (POPG, anionic) vesicles.
