Allosteric regulation of phosphoenolpyruvate carboxylase (PEPC) controls the metabolic flux distribution of anaplerotic pathways. In this study, the feedback inhibition of Corynebacterium glutamicum PEPC was rationally deregulated, and its effect on metabolic flux redistribution was evaluated. Based on rational protein design, six PEPC mutants were designed, and all of them showed significantly reduced sensitivity toward aspartate and malate inhibition. Introducing one of the point mutations (N917G) into the ppc gene, encoding PEPC of the lysine-producing strain C. glutamicum LC298, resulted in ∼37% improved lysine production. In vitro enzyme assays and (13)C-based metabolic flux analysis showed ca. 20 and 30% increases in the PEPC activity and corresponding flux, respectively, in the mutant strain. Higher demand for NADPH in the mutant strain increased the flux toward pentose phosphate pathway, which increased the supply of NADPH for enhanced lysine production. The present study highlights the importance of allosteric regulation on the flux control of central metabolism. The strategy described here can also be implemented to improve other oxaloacetate-derived products.

Japanese encephalitis virus (JEV) is a significant flavivirus that poses a threat to public health, as it induces encephalitis in humans and reproductive disorders in sows. We have recently identified that zinc finger protein 33B (ZNF33B) is required for JEV infection by CRISPR-based functional genomic screening, yet the precise functions and mechanisms are not fully comprehended. In this study, ZNF33B was found to be involved in JEV infection, wherein it bound with JEV RNA to enhance its stability during replication.

Soybean meal (SBM) contains many bioactive compounds, such as isoflavones, which possess anti-inflammatory and anti-oxidative properties that may provide nutritional intervention to pigs infected with porcine reproductive and respiratory syndrome virus (PRRSv). The disease results in abortions, stillborn piglets, and overall impairs reproductive success in sows. Today, there are no data available on feeding SBM to sows infected with PRRSv to mitigate the negative impacts of PRRSv on sow and litter performance.

Antigen-binding proteins, such as nanobodies, modified with functional small molecules hold great potential for applications including imaging probes, drug conjugates, and localized catalysts. However, traditional chemical labeling methods that randomly target lysine or cysteine residues often produce heterogeneous conjugates with limited reproducibility. Conventional site-specific conjugation approaches, which typically modify only the N- or C-terminus, may also be insufficient to achieve the desired functionalities.

These enzymes have two flavin cofactors (FAD and FMN) that each interact with a specific reductant/oxidant substrate/product.
