Article Abstract

Recombinant adeno-associated viruses (rAAVs) that can cross the blood-brain-barrier and achieve efficient and stable transvascular gene transfer to the central nervous system (CNS) hold significant promise for treating CNS disorders. However, following intravascular delivery, these vectors also target liver, heart, skeletal muscle, and other tissues, which may cause untoward effects. To circumvent this, we used tissue-specific, endogenous microRNAs (miRNAs) to repress rAAV expression outside the CNS, by engineering perfectly complementary miRNA-binding sites into the rAAV9 genome. This approach allowed simultaneous multi-tissue regulation and CNS-directed stable transgene expression without detectably perturbing the endogenous miRNA pathway. Regulation of rAAV expression by miRNA was primarily via site-specific cleavage of the transgene mRNA, generating specific 5' and 3' mRNA fragments. Our findings promise to facilitate the development of miRNA-regulated rAAV for CNS-targeted gene delivery and other applications.

Proliferative retinopathy is a leading cause of irreversible blindness in humans; however, the molecular mechanisms behind the immune cell-mediated retinal angiogenesis remain poorly elucidated. Here, using single-cell RNA sequencing in an oxygen-induced retinopathy (OIR) model, we identified an enrichment of sorting nexin (SNX)-related pathways, with SNX3, a member of the SNX family that is involved in endosomal sorting and trafficking, being significantly upregulated in the myeloid cell subpopulations of OIR retinas. Immunostaining showed that SNX3 expression is markedly increased in the retinal microglia/macrophages of mice with OIR, which is mainly located within and around the neovascular tufts.

Abscisic Acid and Calcium Signals Convergently Regulate Sugar Accumulation by Orchestrating the SRK2A/CIPK6-ABI5-TST2 Module in Citrus.

Abscisic acid (ABA) and calcium respectively work as crucial plant hormones and second signalling molecules in the regulation of fruit development and quality formation, including the sugar content and flavour quality. However, the regulatory mechanisms of fruit sugar accumulation arising from the interaction between ABA and calcium have not yet been fully elucidated. Here, we show that the application of ABA or calcium enhances sugar accumulation in sweet orange (Citrus sinensis) fruit, accompanied by upregulation of the expression level of tonoplast sugar transporter 2 (CsTST2), which mediates the transport of sugars into the vacuole.

Patients with Dravet syndrome (DS) present with severe, spontaneous seizures and ataxia. While most patients with DS have variants in the sodium channel Nav1.1 α subunit gene, SCN1A, variants in the sodium channel β1 subunit gene, SCN1B, are also linked to DS.

Organ initiation is often driven by extracellular signaling molecules that activate precursor cells competent to receive and respond to a given signal, yet little is known about the dynamics of competency in space and time during development. Teeth are excellent organs to study cellular competency because they can be activated with the addition of a single signaling ligand, Ectodysplasin (Eda). To investigate the role of Eda in tooth specification, we generated transgenic sticklebacks and zebrafish with heat shock-inducible Eda overexpression.
