Paraoxonase (PON) is a high-density lipoprotein-associated enzyme that prevents low-density lipoprotein oxidation. PON proteins, localized in the seminiferous tubules and in spermatozoa, have been implicated in the pathogenesis of male infertility. In the present study, we sought to explore the contribution of the PON gene variants to sperm parameters. One hundred twenty oligospermic and 170 normozoospermic men were examined during infertility investigation. DNA was extracted from spermatozoa, and the PON1(L/M) 55, PON1(Q/R) 192, and PON2(S/C) 311 polymorphisms were genotyped by polymerase chain reaction and digestion with restriction enzymes. The analysis revealed that oligospermic men presented PON1 55L/L, PON1 192Q/Q, and PON2 311S/S genotypes less frequently than normozoospermic men (P < .01, P < .01, and P < .001, respectively), whereas the PON1 55M, PON1 192R, and PON2 311C alleles were significantly increased in oligospermic men (P < .004, P < .008, and P < .008, respectively). The presence of PON1 55L allele was associated with higher sperm motility in oligospermic men (P < .001), in normozoospermic men (P < .01), and in the total study population (P < .01), and the PON1 192Q allele was associated with higher sperm motility in oligospermic men (P < .01), in normozoospermic men (P < .04) and in the total study population (P < .03). On the other hand, the PON2 311S was associated with higher sperm concentration in oligospermic men (P < .03), in normozoospermic men (P < .008), and in the total study population (P < .001). In our series, the PON1 55M and PON1 192R alleles were associated with decreased sperm motility whereas the PON2 311C allele was associated with decreased concentration, supporting the significance of PON genes in semen quality.

Background: Cryopreservation can lead to sperm damage, which is why incorporating extracellular vesicles derived from normal semen can help protect against the damage caused by freezing.
Materials And Methods: Extracellular vesicles were obtained from normozoospermic semen samples (n = 15), and their effects on the cryopreservation process were studied in three groups: the control group (A, n = 6), the extracellular vesicles treated before freezing group (B, n = 6), and the extracellular vesicles treated after freezing group (C, n = 6). Characteristics of sperm after thawing, including morphology, viability, motility, total antioxidant capacity (TAC), the rate of apoptosis, mitochondrial membrane potential (MMP), and the DNA fragmentation index (DFI), as well as the expression levels of the BAX, BCL, CRISP, and KRAS genes, were examined.

Background: PON3 is associated with high-density lipoprotein (HDL) and plays a protective role against oxidative damage to low-density lipoprotein (LDL). The purpose of the current study was to investigate the effect of semen paraoxonase-3 (PON3) concentration on sperm parameters and DNA fragmentation in both fresh or post-thaw semen.
Methods: This study analyzed PON3 levels in 30 normospermic semen samples obtained from men who attended Dezful Infertility Center and provided informed consent.

Extracellular vesicles (EVs) have been proven to have a crucial role in intercellular communication and have attracted significant attention in the physiology of reproduction because of their multiple functions in physiological processes essential for reproduction including gametogenesis, fertilization and embryo-endometrial cross-talk. Although EVs from the male reproductive tract have been extensively studied for their role in sperm maturation, research on female reproductive tract-derived EVs in humans is still emerging and supported by only a few studies to date. In vitro study was performed using spermatozoa from normozoospermic men and EVs isolated from follicular fluid (FF-EVs), cervicovaginal fluid collected 2 and 7 days after the LH surge (CVF-EVs LH + 2 and LH + 7, respectively) and spent medium of decidualized (dESCs-EVs) and non-decidualized (eESCs-EVs) endometrial stromal cells from healthy women of reproductive age.
