We have developed a highly sensitive single-molecule clonal amplification method called dual primer emulsion PCR (DPePCR) for next-generation DNA sequencing. The approach is similar in concept to standard emulsion PCR; however, in DPePCR both primers are attached to the beads, therefore following PCR amplification, both strands of the PCR products are attached to the beads. The ends of each strand can be freed for analysis by restriction digestion of the bridged PCR fragments, which allows efficient paired-end sequencing of fragment libraries.

Here we investigate the effects and mechanisms of eicosapentaenoic acid (EPA) in regulating chondrocyte mechanics during inflammation in the progression of osteoarthritis (OA). Distinguishing real biological variation in the form of single-nucleotide variants (SNVs) from errors is a major challenge for genome sequencing technologies. This is particularly true in settings where SNVs are at low frequency such as cancer detection through liquid biopsy, or human somatic mosaicism. State-of-the-art molecular denoising approaches for DNA sequencing rely on duplex sequencing, where both strands of a single DNA molecule are sequenced to discern true variants from errors arising from single stranded DNA damage.

Melanoma is the most aggressive type of skin cancer and has very high rates of mortality. The primary objective of this study was to fabricate core-shell nanofibers as a drug delivery system using the coaxial electrospinning technique, which provides some distinct features. Polycaprolactone (PCL)-chitosan (CS)/polyvinyl alcohol (PVA) core-shell nanofibers embedded with Pioglitazone hydrochloride-loaded Nanoemulsions (PIO NEs), a general anti-diabetic drug, and Gemcitabine hydrochloride (GEM), a chemotherapeutic agent, were prepared to investigate the effects of combination of GEM and PIO against A375 melanoma skin cancer cells in vitro.
