The ability to complete DNA replication as replisomes converge has recently been shown to be a highly-regulated, multi-enzymatic process. Converging forks also are likely to generate unique supercoiled, tangled, or knotted substrates. These structures are typically resolved by one of the four topoisomerases encoded by Escherichia coli.

Overcoming resistance to radiotherapy remains a significant challenge in breast cancer management. A one-step coordinated synthesis of BODIPY-integrated photodynamic nanozymes (FZBNPs) that facilitate an orthogonal catalytic cascade for radiotherapy potentiation is presented. The engineered FZBNPs simultaneously alleviate tumor hypoxia through catalase-mimetic oxygen (O) generation and amplify reactive oxygen species (ROS) production via peroxidase-like activity, synergizing with BODIPY-mediated singlet oxygen (O) generation under 660 nm light irradiation.

Disposable electrochemical aptasensors (DEAs) hold significant promise for different analyte detection across diverse fields, due to inherent advantages of rapid response, portability, low cost, and high sensitivity. This review systematically examines the design strategies, signal amplification methodologies, and recent advances in DEAs in the fields of environmental analysis, food safety monitoring, and medical diagnostics. Specifically, it critically evaluates construction strategies for screen-printed electrodes (SPEs) and paper-based electrodes, including substrate selection, ink formulations, and key fabrication techniques such as screen printing, inkjet printing, deposition methods, and direct-writing technologies.

In-field molecular diagnostics of plant pathogens are critical for crop disease management and precision agriculture, but tools are still lacking. Herein, we present a bioluminescent molecular diagnostic assay capable of detecting viable pathogens directly in minimally processed plant samples, enabling rapid and precise in-field crop disease diagnosis. The assay, called bioluminescent craspase diagnostics (BioCrastics), leverages newly discovered RNA-activated protease of CRISPR (Craspase) with enzymatic luminescence to generate a cascaded amplification, thus bypasses nucleic acid purification and amplification while achieving sub-nanogram sensitivity for fungal pathogens.

Background: In neuroblastoma (NB), urinary catecholamine excretion and ImIBG avidity-depending on tumor enzymatic activity and norepinephrine transporter expression, respectively-are diagnostic standards.
