A well known fibrous protein, fibronectin was successfully immobilized onto well-fabricated nanopatterns of polystyrene (PS) substrates. The nanopatterns with finely-defined physical dimensions were obtained by nanoimprinting lithography (NIL) using anodized aluminum oxide (AAO) molds of which the pore sizes were controlled in nanometer scale; 45 nm, 250 nm and 410 nm for the widths and 110 nm and 500 nm for the pitches. Discrete changes of the surface images before and after the protein-adsorption on the patterns were demonstrated by scanning electron microscope (SEM), and the effect of the patterned features on the immobilization has been investigated and discussed.

Platelet integrins, together with other platelet receptors, are known to control hemostasis and thrombosis but also metastatic progression. However, the impact of their exclusive but combined deficiency has never been tested in these processes. In a PF4Cre-β1/β3 mouse strain, we found that platelets were exclusively depleted of all integrins.

Integrin α5β1 is crucial for cell attachment and migration in development and tissue regeneration, and α5β1 binding proteins can have considerable utility in regenerative medicine and next-generation therapeutics. We use computational protein design to create de novo α5β1-specific modulating miniprotein binders, called NeoNectins, that bind to and stabilize the open state of α5β1. When immobilized onto titanium surfaces and throughout 3D hydrogels, the NeoNectins outperform native fibronectin (FN) and RGD peptides in enhancing cell attachment and spreading, and NeoNectin-grafted titanium implants outperformed FN- and RGD-grafted implants in animal models in promoting tissue integration and bone growth.

Matrix Metalloproteinases (MMPs) belong to a family of proteolytic enzymes that degrade extracellular matrix components, such as collagen, elastin, laminin, and fibronectin. They also play a part in tissue remodeling by cleaving and rejoining the tissue proteins. Cancer, neurodegenerative disorders, cardiovascular diseases, arthritis, and chronic inflammatory conditions are just some of the diseases that can start or get worse when different MMPs are not working properly.
