Background: Diabetic nephropathy (DN), a serious diabetic complication, currently has limited treatment options. Yulan Jiangtang capsules (YL) are a clinically approved traditional Chinese medicine formula for glycemic control and diabetes-related complications. Nevertheless, the underlying mechanisms of their therapeutic effects remain incompletely elucidated.

Single-cell RNA sequencing and CRISPR screening enable high-throughput analysis of genetic perturbations at single-cell resolution. Understanding combinatorial perturbation effects is essential but challenging due to data sparsity and complex biological mechanisms. We present GPerturb, a Gaussian process-based sparse perturbation regression model designed to estimate gene-level perturbation effects.

Background: Bone marrow mesenchymal stem cells (BMSCs) exhibit therapeutic potential for osteoporosis through their differentiation into osteoblasts. Here, we investigated the role and mechanism of lymphoid enhancer-binding factor 1 (LEF1) in regulating osteogenic differentiation of human BMSCs (hBMSCs).

Methods: hBMSCs were exposed to the specific medium to induce their osteogenic differentiation.

The aim of this study is to investigate the regulation of Kaixin San-medicated serum(KXS-MS) on autophagy induced by Aβ_(25-35) in SH-SY5Y cells. The SH-SY5Y cell model of Aβ_(25-35)(25 μmol·L~(-1))-induced injury was established, and different concentrations of KXS-MS were added into the culture media of cells, which were then incubated for 24 h. Cell viability was measured by the methyl thiazolyl tetrazolium(MTT) assay.

Article Synopsis
  • GPTON uses GPT-4 to turn structured knowledge, like ontologies, into verbal descriptions, improving how language models handle complex data.
  • The tool effectively annotates over 68% of gene sets in the best predictions, proving its accuracy in text and ontology analysis.
  • Evaluations show GPTON's strength, suggesting it could greatly enhance biomedical research beyond just gene annotation tasks.

The identification of tumor-specific molecular dependencies is essential for the development of effective cancer therapies. Genetic and chemical perturbations are powerful tools for discovering these dependencies. Even though chemical perturbations can be applied to primary cancer samples at large scale, the interpretation of experiment outcomes is often complicated by the fact that one chemical compound can affect multiple proteins.

Humans show micro-expressions (MEs) under some circumstances. MEs are a display of emotions that a human wants to conceal. The recognition of MEs has been applied in various fields.

Developing novel drug formulations and progressing them to the clinical environment relies on preclinical in vitro studies and animal tests to evaluate efficacy and toxicity. However, these current techniques have failed to accurately predict the clinical success of new therapies with a high degree of certainty. The main reason for this failure is that conventional in vitro tissue models lack numerous physiological characteristics of human organs, such as biomechanical forces and biofluid flow.

Tiny changes in the mass of the sensor in a quartz crystal microbalance with dissipation monitoring (QCM-D) can be observed. However, the lack of specificity for target species has hindered the use of QCM-D. Here, molecularly imprinted polymers (MIPs) were used to modify a QCM-D sensor to provide specificity.
