Background: Variants of uncertain significance (VUS) represent a major diagnostic challenge in the interpretation of genetic testing results, particularly in the context of inborn errors of immunity such as severe combined immunodeficiency (SCID). The inconsistency among computational prediction tools often necessitates expensive and time-consuming wet-lab analyses.
Objective: This study aimed to develop disease-specific, multi-class machine learning models using in silico scores to classify SCID-associated genetic variants and improve the interpretation of VUS.

The wrist extends anatomically and functionally from the distal forearm to the metacarpal bones and encompasses the entire carpal region. The large number of bones and joints involved are stabilized in a highly complex system of extrinsic and intrinsic ligaments, which enables a variety of different movements and stability in all positions for the best possible use of the hands. While injuries to individual ligaments or joints, most commonly the scapholunate (SL) ligament, can cause major problems the injuries associated with perilunate dislocations and dislocated fractures represent one of the greatest challenges in hand surgery.

Balancing antibacterial efficacy, mechanical integrity, and biocompatibility remains a critical challenge in drug release systems for wound dressings. Many antimicrobial agents exhibit inherent cytotoxicity, compromising cell viability and tissue compatibility. To address this, an Absorbable Gelatine Sponge was synthetised based on high-viscosity hydroxypropyl methylcellulose (HPMC K100M) and loaded with silver citrate nanorods (AgCit), which confine silver nanoparticles to enable controlled ion release.
