The transcription factor signal transducer and activator of transcription (STAT)4 is a potential target for autoimmune diseases, such as inflammatory bowel disease, multiple sclerosis, rheumatoid arthritis, and diabetes mellitus. p-Biphenyl phosphate is reported as an inhibitor of the STAT4 Src homology 2 domain, and it is developed to the phosphonate-based inhibitor Stafori-1. Herein, structure-activity relationships of p-biaryl phosphates against STAT4 and their selectivity profiles against other STAT proteins are reported.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by joint inflammation. This study aimed to use the sphingosine 1-phosphate receptor 1 (S1PR1) targeted tracer [F]TZ4877 with PET/CT to assess synovial inflammation in a collagen-induced arthritis (CIA) mouse model. [F]TZ4877 and [F]FDG PET/CT imaging were performed on RA ( = 6) and control ( = 6) mice.

Background: Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) are increasingly used in management of type 2 diabetes mellitus (T2DM) and obesity. Beyond glycemic control, these agents may influence orthopaedic outcomes. This study aimed to assess the relationship between preoperative GLP-1 RA use and postoperative complications in T2DM patients undergoing operative ankle fracture repair.
