Megalencephalic leukoencephalopathy with subcortical cysts (MLC) is a rare leukodystrophy characterized by early-onset macrocephaly, white matter edema, seizures, and motor and cognitive decline. Missense mutations to hepatic and glial cell adhesion molecule (hepaCAM), also known as GlialCAM, are responsible for approximately twenty-five percent of MLC cases. HepaCAM is highly enriched in astrocytes and plays important roles in astrocyte territory establishment, gap junction coupling, branching organization, synaptic function, and development of the gliovascular unit.

Proximity-inducing modalities that co-opt cellular pathways offer new opportunities to regulate oncogenic drivers. Inspired by the success of proximity-based chimeras in both intracellular and extracellular target space, here we describe the development of LYsosome Membrane TArgeting Chimeras (LYMTACs) as a small molecule-based platform that functions intracellularly to modulate the membrane proteome. Conceptually, LYMTACs are heterobifunctional small molecules that co-opt short-lived lysosomal membrane proteins (LMPs) as effectors to deliver targets for lysosomal degradation.

Cyanobacteria have an inner and outer cell membrane enclosing the periplasm and cell wall and an additional set of internal membranes (called the thylakoid membranes) enclosing the thylakoid lumen. Teixeira et al. present UltraID-light-inducible protein aggregation (UltraID-LIPA), a technique that combines optogenetic induction of α-synuclein aggregation with proximity-based proteomics. This system enables high-resolution capture of early aggregation events in live cells and implicates known and novel endolysosomal proteins, offering a robust framework for dissecting early pathogenic mechanisms in synucleinopathies and guiding future innovations.
