Sorafenib, a clinically approved multityrosine kinase inhibitor, exhibits poor aqueous solubility, which limits its bioavailability and therapeutic efficacy. In this study, we introduce a solvent-directed self-assembly strategy to modulate the nanostructure of sorafenib without the use of external carriers or complex formulation techniques. In pure water, sorafenib forms large lamellar aggregates, whereas in 30% methanol-water mixtures, it self-assembles into uniform spherical particles approximately 450 nm in diameter. These spherical particles exhibit markedly enhanced cellular uptake (∼80%) and cytotoxicity (up to 60% at 40 μM) in HepG2 liver cancer cells compared to the poorly active lamellar form. In vivo pharmacokinetic analysis further reveals faster absorption, biphasic plasma distribution, and prolonged circulation of the spherical particle forms. These findings demonstrate that solvent-driven nanostructure modulation can serve as a simple yet effective strategy to improve the solubility-limited pharmacological performance of small-molecule drugs like sorafenib.

A number of techniques have been developed to measure the three-dimensional trajectories of protists, which require special experimental set-ups, such as a pair of orthogonal cameras. On the other hand, machine learning techniques have been used to estimate the vertical position of spherical particles from the defocus pattern, but they require the acquisition of a labelled dataset with finely spaced vertical positions. Here, we describe a simple way to make a dataset of images labelled with vertical position from a single 5 min movie, based on a tilted slide set-up.

The protocol presented here enables the quantification of microplastics (MPs) as small as ~1 µm in diameter, accurate identification of polymer types, and estimation of particle volume, critically allowing for the calculation of MP mass. Representative results from samples collected in the Great South Bay (GSB), NY, showed that particles within the 1-6 µm equivalent spherical diameter (ESD) range were the most abundant, with approximately 75% of particles measuring less than 5 µm. Notably, the pre-sieving step failed to yield any particles larger than 60 µm, suggesting that large MPs were rare at the coastal sites sampled.
