Purpose: Pretreatment specimens from patients treated on the I-SPY2 neoadjuvant breast cancer trial were studied to identify prespecified biomarkers associated with response to the regimen of paclitaxel, the anti-type I insulin-like growth factor receptor (IGF-1R) antibody ganitumab, and metformin (PGM) followed by doxorubicin and cyclophosphamide (AC) compared with control therapy (paclitaxel followed by AC). The primary endpoint of this trial is pathologic complete response (pCR).
Experimental Design: One hundred six patients treated with PGM and 119 contemporary controls were evaluated using laser capture microdissection and reverse-phase protein array to evaluate 32 prespecified potential predictive biomarkers in the IGF-1R pathway and 109 additional exploratory endpoints.
Results: Total levels of IGF-1R were poorly correlated with phosphorylated IGF-1R/insulin receptor (IR). Higher levels of phosphorylated IGF-1R/IR were associated with an increased likelihood of obtaining pCR, especially in the hormone receptor (HR)-positive subgroup. Markers of immune response also showed an association with pCR but differed between HR+ and HR- subgroups. In HR- tumors, phospho-STAT1 Y701 and low levels of phospho-p27 associated with pCR. These relationships were not observed in patients treated with control chemotherapy.
Conclusions: Activation status of IGF-1R/IR associated with increased pCR to PGM in HR+ breast cancers. Immune activation markers were also associated with response in HR+ and HR- subgroups. Thus, IGF-1R may directly regulate tumor biology and associate with immune response to therapy.

Proliferative retinopathy is a leading cause of irreversible blindness in humans; however, the molecular mechanisms behind the immune cell-mediated retinal angiogenesis remain poorly elucidated. Here, using single-cell RNA sequencing in an oxygen-induced retinopathy (OIR) model, we identified an enrichment of sorting nexin (SNX)-related pathways, with SNX3, a member of the SNX family that is involved in endosomal sorting and trafficking, being significantly upregulated in the myeloid cell subpopulations of OIR retinas. Immunostaining showed that SNX3 expression is markedly increased in the retinal microglia/macrophages of mice with OIR, which is mainly located within and around the neovascular tufts.

S100 protein family members S100A8 and S100A9 function primarily as a heterodimer complex (S100A8/A9) in vivo. This complex has been implicated in various cancers, including gastric cancer (GC). Recent studies suggest that these proteins play significant roles in tumor progression, inflammation, and metastasis.

To compare the efficacy of using bone marrow mesenchymal stem cell (BM-MSC) exosomes and injectable platelet rich fibrin (i-PRF) on the submandibular salivary glands (SMGs) of aged albino rats in restoring salivary gland structure and function. A total of 40 healthy male albino rats were used, two for obtaining the BM-MSCs, 10 for i-PRF preparation and seven adult rats (6-8 months old) represented the control group (Group 1). The remaining 21 rats were aged (18-20 months old) and divided into three groups of seven rats each; (Group 2): received no treatment, (Group 3): each rat received a single intraglandular injection of BM-MSC exosomes (50 μg/kg/dose suspended in 0.

Background: Colorectal cancer (CRC) remains one of the leading causes of cancer-related mortality worldwide. The tumor microenvironment (TME), particularly the interactions between endothelial cells and cancer-associated fibroblasts (CAFs), plays a pivotal role in promoting tumor growth, angiogenesis, oxidative stress, and therapy resistance. The HUVEC-fibroblast co-culture model closely mimics stromal-endothelial interactions observed in CRC, enabling mechanistic insights not achievable in monocultures.

Objective: To validate and assess clinical efficacy of a prognostic model for predicting severe acute pancreatitis (SAP) based on inflammatory markers (IL-6, ΔIL-22), thromboelastography parameters (K-time) and the BISAP score.
Material And Methods: A prospective observational cohort study enrolled 181 patients with acute pancreatitis. Serum IL-6 and IL-22 were measured in 24 and 48 hours after clinical manifestation, respectively.
