Measurable residual disease (MRD) is a strong prognostic factor in acute myeloid leukaemia (AML). Next-generation sequencing (NGS) offers promise but must distinguish true signal from background. We assessed MRD in 98 adult AML patients in first complete remission after intensive chemotherapy using a duplex unique molecular identifier (UMI)-based NGS capture panel. Error reduction analysis showed up to a 20-fold decrease in artefactual calls versus conventional sequencing. Linearity studies with serial dilutions confirmed accurate quantification down to 0.01% variant allele frequency. In this cohort, NGS-MRD positivity did not significantly affect overall survival (OS) or relapse-free survival (RFS) after one course of chemotherapy. However, NGS-MRD positivity >0.1%, excluding DNMT3A, TET2, ASXL1, IDH1 and IDH2 mutations, was significantly associated with inferior outcomes after two courses (OS: hazard ratio [HR] = 3.04, p = 0.0173; RFS: HR = 2.83, p = 0.0097). Combining multiparameter flow cytometry (MFC-MRD) with NGS-MRD identified a double-positive subgroup with particularly poor outcomes after the first course (OS: HR = 7.98, p < 0.001; RFS: HR = 7.87, p < 0.001). These findings underscore that duplex UMI-based NGS is a sensitive, quantitative approach for MRD assessment in AML, offering prognostic information complementary to MFC-MRD.

Carbon dioxide enhanced oil recovery (CO-EOR) is widely used for carbon capture, utilization, and storage in Chinese oilfields, but part of injected CO returns with produced oil, reducing carbon-reduction efficiency. Bioconverting this CO to methane energy by methanogens benefits the technology, yet on-site high-efficiency conversion meeting natural-gas grid standards remains challenging. This study used a newly-designed triple-tank bioreactor to investigate CO-to-methane conversion and methanogenic kinetics of Methanococcus maripaludis.

The correlation between Pb species formation and bioaccessibility in alkaline, smelter-impacted soil co-contaminated with other toxic trace elements after treatment with phosphorus-containing amendments was investigated. The soil was collected near a former copper smelter, El Paso, Texas. It contained Pb (3200 ± 142 mg kg), As (254 ± 14 mg kg), and Cd (110 ± 8 mg kg).

The susceptibility to emotional contagion has been psychometrically addressed by the self-reported Emotional Contagion Scale. With the present research, we validated a German adaptation of this scale and developed a mimicry brief version by selecting only the four items explicitly addressing the overt subprocess of mimicry. Across three studies (N1 = 195, N2 = 442, N3 = 180), involving various external measures of empathy, general personality domains, emotion recognition, and other constructs, the total German Emotional Contagion Scale demonstrated sound convergent and discriminant validity.

Purpose: To discuss the technique and outcome of what the authors called the "flap-in-flap" technique and report its safety as a procedure for correction of post-laser in situ keratomileusis (LASIK) myopic regression.
Methods: Seven eyes of 4 patients were included in this study. All patients had previously undergone LASIK for compound myopic astigmatism using the Moria M2 micro-keratome (Moria) 8 to 12 years prior to presentation.
