Article Abstract

Hemoglobin A (HbA) plays a role in the diagnosis and assessment of glycemic control in patients with diabetes. However, few studies have focused on the preparation methods and analytical performance evaluation of different formulations of mobile-phase buffers for HbA assays. In this study, mobile-phase buffers of new formulations were prepared for the measurement of HbA. Following the Clinical and Laboratory Standards Institute (CLSI) guidelines, the analytical performance of newly developed mobile-phase buffers was evaluated. The CVs of the intra- and inter-assay precision were less than 0.94%, the RDs of accuracy were less than ± 3.00%, and the carryover was 0.00%. The measured HbA values were linear from 4.50% to 16.80% (26.00-160.00 mmol/mol). A comparison of newly developed and commercial mobile-phase buffers revealed that the Pearson regression equation was Y = 0.9862x + 0.0888 (R = 0.9990), and the Bland-Altman mean difference between the two mobile-phase buffers was -0.02833% (95% CI: -0.1853% to 0.1278%). The chromatography column had a lifetime of over 7000 tests. The reference interval was 4.30%-6.30% (23.00-45.00 mmol/mol). Stability was acceptable within 24 months. The newly developed mobile-phase buffers demonstrated good analytical performance (i.e., precision, linearity, accuracy, carryover, reference interval, the number of chromatography column runs, and stability).

The composition of the injection solvent is a critical, yet often underestimated, parameter in liquid chromatography-tandem mass spectrometry (LC-MS/MS). This study systematically evaluates the influence of injection solvent on the analysis of 90 pesticides by comparing mixtures of acetonitrile (ACN) with water and buffered mobile phase A (5 mM ammonium formate, 0.1% formic acid) across various ratios (10/90 to 50/50, v/v).

With the growing demand for high-throughput analyses that can detect diverse molecules with varying physicochemical properties in shorter times, researchers are increasingly focused on developing or modifying analytical methods. This is particularly relevant in the food, pharmaceutical/nutraceutical, cosmetic, agricultural, and environmental industries. This study aimed to modify, establish, and validate a high-performance liquid chromatography method with ultraviolet detection (HPLC-UV) for the simultaneous determination of disodium guanylate (GMP) and disodium inosinate (IMP) in mushrooms, using as a model.

A simple, precise, and stability-indicating reverse-phase high-performance liquid chromatography method was developed and validated for the simultaneous estimation of dapagliflozin, linagliptin, and metformin hydrochloride in fixed-dose combination tablets. Chromatographic separation was achieved using a Phenomenex Luna C18 column (250 × 4.6 mm, 5 μm) with a mobile phase consisting of acetonitrile and phosphate buffer (pH 6.

Although chlorpromazine is primarily used in psychiatry, it has been shown since its introduction to influence the course of neoplastic diseases. According to the strategy of drug repurposing, chlorpromazine has been successfully tested for its potential antitumor effects on multiple cancer cell lines. This effect is consistent with the overlap of molecular pathways observed for years between schizophrenia and cancer.
