Antibody-drug conjugates (ADCs) represent a promising therapeutic approach in gynecologic cancers, particularly ovarian and cervical malignancies. Agents such as mirvetuximab soravtansine, and tisotumab vedotin, targeting folate receptor alpha and tissue factor, respectively, reported clinical efficacy in patients with limited options. However, their use is associated with ocular toxicities, including keratopathy, blurred vision, and dry eye, which may impact adherence and quality of life.

Radiotherapy (RT) is a mainstay of cancer treatment but is limited by tumor resistance and off-target tissue damage, often mediated by therapy-induced cellular senescence. Here, we developed a "one-two punch" nanodrug, Lipo@ABT263@Au, that integrated a senolytic agent (ABT-263) with a gold-shelled liposome for radiosensitization and sustained drug release. High-throughput screening and transcriptomic analysis identified senescence as a key RT-induced vulnerability.

Management of recurrent adenoid cystic carcinoma (ACC) in elderly patients remains challenging due to comorbidities, functional impairments, and anatomically complex tumor locations that complicate surgical access and increase operative risk. The ZAP-X Gyroscopic Radiosurgery System (ZAP Surgical Systems, Inc., San Carlos, CA, USA) offers a highly precise, non-invasive treatment modality, potentially suitable for salvage therapy in previously irradiated fields and in medically inoperable patients.
