Article Abstract

Photolysis of 3-phenyl- and 3-(3-pyridyl)-sydnones in Ar matrices is known to furnish Earl's bicyclic lactone (bicyclo[2.1.0]oxadiazolone) followed by the elimination of CO and the formation of carbodiimides . Laser photolysis of 3-arylsydnones at 266 nm in an MTHF matrix at 5 K generates triplet arylnitrenes , which are characterized by their zero-field splitting parameters determined from their electron spin resonance (ESR) spectra. The potential routes from nitrile imines , diazirines , imidoylnitrenes , and carbodiimides to arylnitrenes are investigated computationally at the M06-2X/6-311+G(d,p) level. Imidoylnitrenes are the key intermediates in the matrix photolyses of sydnones as well as tetrazoles, whereby the triplet states of dissociate to a nitrene () and a nitrile, and the open-shell singlet state rearranges to a carbodiimide ; these two processes have nearly the same activation energies of 20-22 kcal/mol.

Nitric oxide donor-mediated inhibition of phosphorylation shows that light-mediated degradation of photosystem II D1 protein and phosphorylation are not tightly linked.

Sustainable Agricultural Systems Laboratory, Building 001, Beltsville Agricultural Research Center, Agricultural Research Service, United States Department of Agriculture, Beltsville, MD 20705-2350, USA.

An outcome of the photochemistry during oxygenic photosynthesis is the rapid turn over of the D1 protein in the light compared to the other proteins of the photosystem II (PS II) reaction center. D1 is a major factor of PS II instability and its replacement a primary event of the PS II repair cycle. D1 also undergoes redox-dependent phosphorylation prior to its degradation.

How to photostabilize molsidomine tablets.

J Pharm Sci

July 2004

Department of Pharmaceutical Technology, Ludwig-Maximilians University, Munich, Germany.

Different methods of photostabilization are presented for the very light sensitive molsidomine tablets. The incorporation of photostabilizers such as light absorber or pigments into the tablets considerably improved the photostability. Nevertheless, photodegradation was still detected after 12 h of intense light stress.

The ICH guideline Q1B for photostability testing gives guidance on the basic testing protocol required to evaluate the light sensitivity and stability of new drugs and products. The choice of the irradiation method, although complying with the guideline demands, may effect test results. High irradiances may shorten testing times, but can lead to enforcement of photodegradation, which was demonstrated for molsidomine tablets.

Particular features of photolabile substances in tablets.

Pharmazie

September 2003

Department of Pharmaceutical Technology, Ludwig-Maximilians-University Munich, Germany.

Nifedipine and molsidomine tablets are extremely photolabile drug preparations, even at cool room light. Compared to solutions the light spectrum responsible for photodegradation is moved towards the long-wavelength range corresponding to the bathochromic shift of light absorption in the solid state. In the case of nifedipine tablets light up to 500 nm, especially the range between 400-420 nm, is degrading.
