This study evaluates and compares four millimeter-wave (mmWave) radio-over-fiber (RoF) frequency multiplexing techniques considering InAs/InP quantum dash (QD) mode-locked lasers (MLLs) for optical carriers. The QD-MLL can generate multiple coherent optical carriers simultaneously with consistent frequency differences. Following heterodyne detection, the radio frequency linewidths of the QD-MLL can be minimized to 2.4 kHz. In this paper, four distinct RoF-based mmWave frequency multiplexing architectures are presented and compared experimentally. It is shown that the error vector magnitude of two 2 GBaud/s 16-QAM mmWave signals can reach as low as 7.1% after demultiplexing for all four techniques.

Amplicon sequencing is a popular method for understanding the diversity of bacterial communities in samples containing multiple organisms as exemplified by 16S rRNA sequencing. Another application of amplicon sequencing includes multiplexing both primer sets and samples, allowing sequencing of multiple targets in multiple samples in the same sequencing run. Multiple tools exist to process the amplicon sequencing data produced via the short-read Illumina platform, but there are fewer options for long-read Oxford Nanopore Technologies (ONT) sequencing, or for processing data from environmental surveillance or other sources with many different organisms.

Multiplex assays of variant effect (MAVEs) provide promising new sources of functional evidence, potentially empowering improved classification of germline genomic variants, particularly rare missense variants, which are commonly assigned as variants of uncertain significance (VUSs). However, paradoxically, quantification of clinically applicable evidence strengths for MAVEs requires construction of "truthsets" comprising missense variants already robustly classified as pathogenic and benign. In this study, we demonstrate how benign truthset size is the primary driver of applicable functional evidence toward pathogenicity (PS3).
