Designing CO-releasing molecules, which store, transport, and release carbon monoxide in the target tissue, has accelerated since scientists revealed that carbon monoxide is one of the transmitters and could be effective in treatment procedures. The most important candidates for this task are metal carbonyl complexes. In this study, [Mn-(CO)(bpy)-L]-PF-type metal carbonyl complexes were synthesized and characterized and the CO-releasing activities of these molecules were investigated. In addition, the optimization and theoretical analysis of the molecules were performed with DFT/TDDFT-based calculation methods. DFT computations at the B3LYP/6-311G-(d,p)/LANL2DZ level were performed to assign vibrational modes and NMR shifts following geometry optimization and confirmation. Moreover, NBO analyses were performed to predict the important electronic interaction that occurred in complexes: the results implied that the biggest contribution could come from the resonance interactions. FMOs analyses indicated that the 2e could be a softer (η= 1.511 eV) and is less stabilized complex via back-donation (Δε = -0.378 eV). Additionally, the interactions of the molecules with HSA, BSA, and DNA were investigated with molecular docking methods, and the binding properties of the manganese complexes were analyzed in vitro with UV-vis spectroscopy against BSA and DNA by the Benesi-Hildebrand method.

Background: Lung cancer remains the leading cause of cancer-related mortality globally, primarily due to late-stage diagnosis, molecular heterogeneity, and therapy resistance. Key biomarkers such as EGFR, ALK, KRAS, and PD-1 have revolutionized precision oncology; however, comprehensive structural and clinical validation of these targets is crucial to enhance therapeutic efficacy.
Methods: Protein sequences for EGFR, ALK, KRAS, and PD-1 were retrieved from UniProt and modeled using SWISS-MODEL to generate high-confidence 3D structures.

This study utilized integrated sensory-guided, machine learning, and bioinformatics strategies identify umami-enhancing peptides from , investigated their mechanism of umami enhancement, and confirmed their umami-enhancing properties through sensory evaluations and electronic tongue. Three umami-enhancing peptides (APDGLPTGQ, SDDGFQ, and GLGDDL) demonstrated synergistic/additive effects by significantly enhancing umami intensity and duration in monosodium glutamate (MSG). Furthermore, molecular docking showed that these umami-enhancing peptides enhanced both the binding affinity and interaction forces between MSG and the T1R1/T1R3 receptor system, thereby enhancing umami perception.

A novel isatin-thiazole-coumarin hybrid and three isatin-hydantoin hybrids were synthesized and assessed for their α-glucosidase and anticholinesterase inhibitory activities. Moreover, their anticancer properties have been observed against the breast cancer cell lines MCF-7 and MDA-MB-231. The coumarin-containing hybrid exhibited the most potent biological activity across all assays.

Mammalian ALOX15 are allosteric enzymes but the mechanism of allosteric regulation remains a matter of discussion. Octyl (-(5-(1-indol-2-yl)-2-methoxyphenyl)sulfamoyl)carbamate inhibits the linoleate oxygenase activity of ALOX15 at nanomolar concentrations, but oxygenation of arachidonic acid is hardly affected. The mechanism of substrate selective inhibition suggests inter-monomer communication within the allosteric ALOX15 dimer complex, in which the inhibitor binding to monomer A induces conformational alterations in the structure of the active site of monomer B.

This research work details the use of a molecular hybridization technique to create a library of four series of hydrazineyl-linked imidazo[1,2-]pyrimidine-thiazole derivatives. The structure of one of the final products, K2, was validated using single-crystal X-ray diffraction. Twenty-six novel hybrid molecules (K1-K26) were synthesized and tested for activity against the H37Rv strain.
