Skin diseases typically lead to the disruption of the functional barrier of human skin, and glucocorticoids (GCs) can effectively treat these conditions through topical administration. However, the molecular mechanisms underlying the uptake of GCs by the skin remain inadequately understood. In this study, we employed molecular dynamics (MD) simulations to investigate the uptake of seven GCs by the stratum corneum (SC). The results indicate that hydrocortisone (HC), halobetasol propionate (HP), fluticasone propionate (FP), and mometasone furoate (MS) are more readily absorbed by the SC. The uptake process for HC can be divided into three distinct stages: contact stage, transformation stage, and absorption stage. Furthermore, our analysis of drug nanoparticle release revealed that electrostatic interactions between HP molecules were more significant than van der Waals interactions; additionally, the free energy associated with the release process of HP was greater than that of HC. To further elucidate how interactions between GCs and SCs influence GC uptake processes, we examined the relationship between 17 HC molecules' interactions with SCs and their depth of penetration within the SC. A predictive model was developed using one-way linear regression based on these findings. In conclusion, this work offers valuable insights into GC uptake mechanisms at a molecular level concerning the SC.

Surface electromyography (sEMG) is the measurement of the electrical activity of muscle and is extensively used in fundamental research and across many applications in health and sport. Conventional surface electrode technology can suffer from poor signal quality, particularly when used outside the laboratory, requires careful skin preparation prior to electrode application, and can be challenging when used for long-term recording. These limitations have challenged the translation of sEMG to widespread clinical application.

Diabetic wounds present persistent challenges due to impaired healing, recurrent infection, oxidative stress, and dysregulated glucose metabolism. Bioinspired polymeric microneedle (MN) patches have emerged as multifunctional platforms capable of penetrating the stratum corneum to deliver therapeutics directly into the dermis, enabling glucose regulation, antimicrobial action, reactive oxygen species (ROS) modulation, and proangiogenic stimulation. Recent experimental evidence has demonstrated that the integration of glucose oxidase-loaded porous metal-organic frameworks, photothermal nanomaterials, and antioxidant hydrogels within dissolvable MNs achieves synergistic bactericidal effects, accelerates collagen deposition, and enhances neovascularization in diabetic wound models.

Recalcitrant palmar common warts pose a considerable challenge in dermatology due to their frequent persistence despite various treatment attempts. The thick stratum corneum of the palms and the constant pressure and friction in this location contribute to their resistance to therapy and a high rate of recurrence. We report the case of a 33-year-old male with a 26-month history of a progressively enlarging palmar wart refractory to extensive conventional therapies, including 18 intermittent sessions of liquid nitrogen cryotherapy administered over the course of his 26-month history, two electrofulguration sessions, and various topical agents.
