Article Abstract

Activating mutations in the epidermal growth factor receptor (EGFR) are key oncogenic drivers across multiple cancers, yet the structural mechanisms by which these mutations promote persistent receptor activation remain elusive. Here, we investigate how three clinically relevant mutations-T790M, L858R, and the T790M_L858R double mutant-reshape EGFR's conformational ensemble and regulatory network architecture. Using multiscale molecular simulations and kinetic modeling, we show that these mutations, particularly in combination, enhance flexibility in the αC-helix and A-loop, favoring activation-competent states. Markov state modeling reveals a shift in equilibrium toward active macrostates and accelerated transitions between metastable conformations. To resolve the underlying coordination mechanism, we apply neural relational inference to reconstruct time-dependent interaction networks, uncovering the mutation-induced rewiring of allosteric pathways linking distant regulatory regions. This coupling of conformational redistribution with network remodeling provides a mechanistic rationale for sustained EGFR activation and suggests new opportunities for targeting dynamically organized allosteric circuits in therapeutic design.

Emerging evidence underscores the regulatory roles of nonamyloidogenic regions in controlling the aggregation dynamics and cytotoxicity of amyloidal proteins, but the mechanism remains unclear. Herein we investigated how flanking sequences modulate the conformational heterogeneity in the p53 238-262 amyloid segment using scanning tunneling microscopy (STM). By comparing the wild-type (wt) and three pathogenic mutations (R248W, R248Q, R249S) in the noncore regions, we reveal that flanking alterations remodel β-sheet aggregates and induce conformational plasticity in β-strand ensembles through the generation of novel conformational substates and selective elimination of existing conformational substates.

The Guided Entry of Tail-Anchored Proteins (GET) pathway ensures accurate targeting of Tail-Anchored proteins (TAs) - a diverse class of membrane proteins - to the endoplasmic reticulum (ER) membrane. In yeast, newly synthesized TAs are captured by Sgt2 and transferred to Get3 for delivery to the ER, where they undergo subsequent membrane insertion. Efficient and protected handoff of hydrophobic TAs from Sgt2 to Get3 is facilitated by the Get4/5 complex, which is thought to act as a scaffold to position TA-bound Sgt2 and Get3 in proximity while trapping Get3 in an ATP-bound conformation necessary for TA binding.

α-1,3-Fucosyltransferase (α-1,3-FucT), the rate-limiting enzyme in the biosynthesis of the human milk oligosaccharide 3-fucosyllactose (3-FL), suffers from limited industrial applicability due to its low catalytic activity and poor thermostability. Herein, we systematically optimized the catalytic performance and thermostability of M32-H10 using computer-aided design coupled with a greedy combinatorial strategy.
