The long non-coding RNA reportedly forms a circular RNA variant (circ). As circ is expressed in various cancers and has been implicated in promoting cancer cell proliferation and tumor progression, it is considered a potential biomarker and therapeutic target. We previously confirmed that circ expression varies according to the Gleason pattern, a morphological indicator of malignancy in prostate cancer. In this study, we assessed the expression of circ using BaseScope assay with prostate cancer tissues and evaluated the correlation with the Grade Group (based on Gleason pattern), an indicator used to morphologically evaluate the degree of malignancy of prostate cancer. The relationship between circ expression and tumor proliferation was evaluated using cells in which circ expression was suppressed using the clustered regularly interspaced short palindromic repeats (CRISPR)/RfxCas13d system. BaseScope assay confirmed that circ expression was significantly higher in Grade Group 2-5 (intermediate- and high-grade groups) than Grade Group 1 (low-grade group). experiments using the CRISPR/RfxCas13d system showed that specific suppression of circ expression resulted in a significant reduction in the number of prostate cancer cells. The results of this study suggest that circ is involved in tumor growth in prostate cancer and may serve as a therapeutic target for moderately and highly malignant prostate cancers that express circ.

Excess testosterone (T) exposure from early to mid-gestation (days 30-90) leads to sexually dimorphic adverse cardiac left ventricular (LV) programming at fetal day 90 (term 147 days). Whether this sexually dimorphic impact is a direct effect of T or reprogramming that persists beyond early fetal life is unknown. We hypothesized that adverse sex-specific cardiac outcomes seen in early fetal life will persist in late gestational fetuses.

Intravascular hemolysis (IVH), a pathological process associated with various conditions, triggers inflammatory responses, yet the key molecular drivers of these responses are poorly defined, particularly within the vasculature. To explore the role of NLRP3 inflammasome- and caspase-1-dependent pathways in IVH-induced vascular dysfunction, we used models of acute and chronic IVH, alongside heme stimulation of endothelial cells, thereby isolating this disease mechanism from its etiological causes. IVH induced rapid inflammatory responses in C57BL/6J mice, including IL-1β release within 15 minutes, and NLRP3-dependent caspase-1 activation in circulating leukocytes.

Background: Calcium (Ca) dysregulation is a hallmark of heart failure, impairing excitation-contraction coupling and contributing to pathological remodeling. The SERCA2a (sarco/endoplasmic reticulum Ca ATPase isoform 2a) mediates Ca reuptake into the sarcoplasmic reticulum (SR) during diastole, but its activity declines in failing hearts. DWORF (dwarf open reading frame), a newly identified cardiac microprotein, enhances SERCA2a activity and improves cardiomyocyte Ca cycling and contractility.
