This study reports the production, statistical optimization, partial purification and characterization of a highly efficient xylanase from a high-altitude soil bacterium, Bacillus altitudinis XYL17, for the enzymatic valorization of agrowaste. A two-step optimization approach combining Plackett-Burman Design (PBD) and Response Surface Methodology (RSM) enhanced xylanase production with minimal resource utilization. The partially purified enzyme exhibited optimal activity at 50-55 °C and pH 6.5, and maintained stability over a broad temperature (5-65 °C) and pH (5.0-9.5) range. Substrate specificity analysis revealed efficient hydrolysis of beechwood xylan (BX) and alkali-extracted corncob xylan (CCX) into major xylooligosaccharides (XOS), achieving a combined yield of 48 % for xylobiose and xylotriose per gram of BX. The XOS displayed antioxidant properties through hypochlorous acid (HOCl) reduction and myeloperoxidase (MPO) inhibition, and supported the growth of probiotic strains Lactobacillus acidophilus MTCC 10703 and Limosilactobacillus fermentum MCC 2759. These findings highlight the potential of B. altitudinis XYL17 as a promising source for industrial xylanase production and establish its xylanase as a robust and versatile biocatalyst for the sustainable conversion of xylan-rich biomass into functional biomolecules, with potential applications in the food and nutraceutical industries.

Functional high risk multiple myeloma (FHRMM) remains a challenging entity with poor outcomes and limited survival, and there is no international consensus on optimal second-line therapeutic strategies in relapsed/refractory patients. In this multicenter real-world retrospective study, we investigated clinical characteristics and outcomes of a total of 62 FHRMM patients previously treated with a first-line daratumumab-based quadruplet regimen or who relapsed within 12 months after frontline autologous stem cell transplantation (ASCT). In our cohort, the overall response rate was 61%, with 42% of patients achieving a very good partial response (VGPR) or better.

The MET receptor tyrosine kinase is a pivotal regulator of cellular survival, motility, and proliferation. Mutations leading to skipping of exon 14 (METΔex14) within the juxtamembrane domain of MET impair receptor degradation and prolong oncogenic signaling, contributing significantly to tumor progression across multiple cancer types. This study explores the economic implications of transport remit management in Poland's international trade landscape, with a particular focus on the operations of a medium-sized Polish forwarding company (Company X). Employing a mixed-methods approach, the research combines quantitative analysis of government datasets, firm-level transaction data, and qualitative insights from a targeted industry survey. The case study of Company X reveals notable reluctance among Polish enterprises to assume transport remit responsibilities, particularly in import operations, due to preferences for foreign partners, limited experience with international logistics, and concerns about administrative complexity.
