Article Abstract

The spectrin family genes play critical roles in cytoskeletal organization and cellular integrity, yet their evolutionary and functional significance in non-classical model organisms remains poorly explored. Here, we systematically identified and characterized spectrin family genes in the silkworm . Genome-wide analysis identified 17 predicted spectrin genes, which were integrated into eight optimized members through transcriptome-guided structural refinement. Multi-species genomic analysis revealed 8, 23, and 24 spectrin family genes in , , and , respectively. Phylogenetic analysis revealed conserved clades across insects and mammals, with gene family expansions in vertebrates. Spatiotemporal expression profiling demonstrated ubiquitous expression of these genes during silkworm development. Population genomic analyses detected strong selection signatures in during domestication and implicated as a candidate gene for silk yield enhancement in Chinese-improved strains (CHN-I). Expression profiles of parental strains and F offspring from a commercial hybrid cross (Jingsong × Haoyue) revealed expression correlating with heterosis in silk yield traits. This study elucidates the characterization and functional relevance of silkworm spectrin genes, providing insights into their roles in domestication and breeding.

Aims: The current therapeutic approach to ischaemic (IsHF) and non-ischaemic (NIsHF) heart failure (HF) mainly overlooks the underlying aetiology owing to a lack knowledge of the differential molecular pathways that contribute to HF with reduced ejection fraction (HFrEF). Alterations in myocardial DNA methylation levels have been identified as potential biomarkers for HF irrespective of its aetiology. Due to the limited availability of cardiac tissues in clinics, our goal is to determine if DNA methylation changes in circulating CD4 T lymphocytes, which are strongly involved in left ventricle remodelling, can help in differentiating IsHF and NIsHF causes among patients with HFrEF and if DNA methylation levels associate with key clinical features.

: Adducins are cytoskeletal proteins essential for membrane stability, actin-spectrin network organization, and cell signaling. Mutations in the genes , , and have been linked to hypertension, neurodevelopmental disorders, and cancer. However, no comprehensive in silico saturation mutagenesis study has systematically evaluated the pathogenic potential and structural consequences of all possible missense mutations in adducins.

is a high confidence autism spectrum disorder (ASD) gene encoding the spectrin-actin scaffold protein Ankyrin B (AnkB). The 220 kDal isoform of AnkB has multiple functions including developmental spine pruning through L1 family cell adhesion molecules (L1-CAMs) and class 3 Semaphorins on dendrites of pyramidal neurons to achieve an appropriate excitatory balance in the neocortex. Molecular modeling employing AlphaFold was used to predict the structure and interactions of AnkB with the cytoplasmic domain of Neuron-glial Related L1-CAM (NrCAM), and with β2-Spectrin.

Autism spectrum disorder (ASD) is a neurodevelopmental impairment that occurs due to mutations related to the formation of the nervous system, combined with the impact of various epigenetic and environmental factors. This necessitates the identification of the genetic variations involved in ASD pathogenesis. We performed whole exome sequencing (WES) in a cohort of 22 Bulgarian male and female individuals showing ASD features alongside segregation analyses of their families.
