Article Abstract

Although posited as an explanation for reproductive endocrine-related mood disorders, differential hormone sensitivity is an elusive concept. In this editorial, we define differential sensitivity, embed it in current understanding of the generation of brain states and discuss its practical utility.

Observing differential spin currents by resonant inelastic X-ray scattering.

Nature

September 2025

National Synchrotron Light Source II, Brookhaven National Laboratory, Upton, NY, USA.

Controlling spin currents, that is, the flow of spin angular momentum, in small magnetic devices, is the principal objective of spin electronics, a main contender for future energy-efficient information technologies. A pure spin current has never been measured directly because the associated electric stray fields and/or shifts in the non-equilibrium spin-dependent distribution functions are too small for conventional experimental detection methods optimized for charge transport. Here we report that resonant inelastic X-ray scattering (RIXS) can bridge this gap by measuring the spin current carried by magnons-the quanta of the spin wave excitations of the magnetic order-in the presence of temperature gradients across a magnetic insulator.

At present there is no metabolic characterization of acute promyelocytic leukemia (APL). Pathognomonic of APL, PML::RARα fusion protein rewires metabolic pathways to feed anabolic tumor cell's growth. All-trans retinoic acid (ATRA) and arsenic trioxide (ATO)-based therapies render APL the most curable subtype of AML, yet approximately 1% of cases are resistant and 5% relapse.

With the approval of the antibody-drug conjugate enfortumab vedotin (EV), NECTIN4 has emerged as a bona fide therapeutic target in urothelial carcinoma (UC). Here, we report the development of a NECTIN4-directed chimeric antigen receptor (CAR) T cell, which exhibits reactivity across cells expressing a range of endogenous NECTIN4, with enhanced activity in high expressors. We demonstrate that the PPARγ pathway, critical for luminal differentiation, transcriptionally controls NECTIN4, and that the PPARγ agonist rosiglitazone primes and augments NECTIN4 expression, thereby increasing sensitivity to NECTIN4-CAR T cell-mediated killing.

Bone morphogenetic proteins (BMPs) are effective for treating various orthopedic conditions and are widely used clinically. However, their therapeutic efficacy is limited in osteoporosis patients. Iron overload represents a key risk factor for osteoporosis, inducing ferroptosis and suppressing the osteogenic differentiation of bone marrow stromal cells (BMSCs).

Given the pivotal role of Flap endonuclease 1 (FEN1) in tumor pathogenesis and progression, the advancement of its activity and inhibitor assays holds significant importance for cancer research and drug screening. Herein, we proposed a convenient, visual and sensitive colorimetric biosensing platform for FEN1 activity detection by integrating the robust signal amplification power of rolling circle amplification (RCA), the target enrichment capability of magnetic beads (MB), and the high efficiency and visualization of urease-mediated litmus test. Based on the significant color transition with a clear response mechanism, quantitative analysis can be achieved by either spectroscopic or smartphone-based detection.
