Article Abstract

Riboswitches are non-coding RNA motifs that regulate gene expression in response to ligand binding. The glycine tandem riboswitch (GTR) is notable because it comprises two distinct glycine aptamers that interact extensively. These inter-aptamer contacts drive conformational changes in the downstream expression platform to control gene expression. Despite extensive studies, the role of glycine and RNA folding pathways in co-transcriptional regulation remains unclear. Here, we integrate single-molecule kinetic analysis, co-transcriptional RNA structure probing, and computational modeling to reveal that the GTR processes multiple molecular inputs sequentially, guided by polymerase pausing. Our findings elucidate its stepwise 5'-to-3' folding pathway and demonstrate how sequential glycine binding to each aptamer, K binding to a kink-turn, non-native RNA folding intermediates, inter-aptamer docking that drives binding site pre-organization, and modulation by the transcription factor NusA collectively orchestrate co-transcriptional gene regulation. These results support a model in which glycine binding cooperativity arises through non-equilibrium mechanisms rather than a classical concerted model.

This study aimed to analyze the amino acid composition and characterize the sequences of collagen peptides from Skipjack tuna bones (TBCPs) by liquid chromatography-tandem mass spectrometry (LC-MS/MS), and further investigate the function and mechanism of action of TBCPs in nonalcoholic fatty liver disease (NAFLD). The results showed that TBCPs contain 16 types of amino acids, among which glycine is the most abundant, and hydrophobic amino acids account for 40.75 %.

Background: Previous studies involving cleanup via conventional solid-phase extraction (SPE) materials to overcome matrix effects for the polar organophosphonate and -phosphinate pesticides glyphosate, glufosinate, ethephon, fosetyl, and their various metabolites often showed limitations due to the existence of various matrix compounds in plant commodities with similar polarity. To overcome existing drawbacks, we utilized the unique selectivity provided by metal oxides as SPE materials. These were exploited in a novel automated online SPE-LC-MS/MS method which allowed analyte-specific trapping in the presence of excessive amounts of matrix compounds as typically contained in extracts of the Quick Polar Pesticides (QuPPe) method.

Renal complications are common following heart transplantation and lead to increased morbidity and mortality. The incidence of renal insufficiency is 10%-40% at 3-7 years after transplantation. This study aims to conduct metabolic profiling of serum samples from heart transplantation recipients to uncover differential metabolites related to renal insufficiency and potential pathogenic mechanisms.

Background: Neurotransmitter disorders are a group of heterogeneous conditions that comprise defects in synthesis, transport, receptor binding, and degradation of neurochemical messengers. These rare disorders range from mild intermittent dystonia to lethal encephalopathies. The natural history and clinical presentation remain far from established.
