Article Abstract

(Cm) has reemerged as a prevalent infectious agent in research mouse colonies. Despite its prevalence and ability to persistently colonize the murine gastrointestinal tract, few studies have evaluated the potential impact of Cm on experimental models of gastrointestinal disease. Studies were conducted to evaluate the impact of Cm on the (Cr), (Tm) and 10 mouse models of intestinal inflammation, as well as on tumorigenesis in the Apc mouse following administration of DSS. Naïve C57BL/6J (B6), B6.129P2- 10 /J ( 10 ) and C57BL/6J-Apc /J (Apc ) mice were infected with Cm by cohousing with chronic Cm-shedding BALB/cJ mice for 2 weeks; controls were cohoused with Cm-free mice. After cohousing, B6 mice (n=8 Cm-infected and free) were infected with (Cr; 10 CFU orally) or (Tm; 200 ova orally). 0 mice (n=8/group with and without [10 CFU/mouse] and with and without Cm) and Apc mice (n=8/group) received 2% DSS for 7 days in drinking water after cohousing. Mice were sacrificed 14-days post-Cr infection, 18-days post-Tm infection, 70-days after cessation of cohousing with 10 mice, and 28-days post-DSS administration to Apc mice. The severity of the cecal and colonic lesions were evaluated and graded using a tiered, semi-quantitative scoring system. Cm infection attenuated colitis associated with Cr (p=0.03), had no effect on Tm associated pathology (p=0.22), worsened colitis in 10 mice in the absence of (p=0.007), and reduced chemically induced colonic tumorigenesis in Apc mice (p=0.004). Thus, Cm colonization differentially impacts several models of intestinal inflammation and tumorigenesis, and the presence of this bacterium in mouse colonies should be considered as a variable in these experimental readouts.

Establishing immune tolerance to gut microbiota and food antigens upon first exposures during early life is essential to prevent inflammatory bowel diseases and food allergy and depends on induction of peripherally induced Rorgt expressing regulatory T (Rorgt+ pTreg) cells. Recent studies have identified a critical role for Rorgt expressing antigen-presenting cells (APC), Thetis cells (TCs), in peripheral regulatory T (pTreg) cell differentiation and tolerance to food and commensal microbes. We present a series of preclinical studies focused on developing in vitro 2D and 3D models for assessing immunogenic factors in preventing infectious diseases. Human peripheral blood mononuclear cells (PBMC) and Calu-3 cell lines (bronchial epithelial cells) were used to develop 2D and 3D models. Peptides: Spike-S1-His (S-His), nucleocapsid-His and adjuvants: human adenovirus five serotype-based viral vector (AdV-D24-ICOSL-CD40L), armed with inducible co-stimulator (ICOSL) and CD40 ligand (CD40L), and a vector lacking these transgenes (AdV5/3) were used due to their effective initial interaction with antigen-presenting cells (APC).
