Article Abstract

Neoantigen-reactive peripheral blood lymphocytes (NeoPBL) are tumor-specific T cells found at ultra-low frequencies in the blood. Unlike tumor-infiltrating lymphocytes (TIL), NeoPBL exist in a favorable less dysfunctional phenotypic state , but their rarity has precluded their effective use as cell therapy. Leveraging knowledge of bona fide neoantigens, we combined high-intensity neoantigen stimulation with bead extraction of neoantigen peptide-pulsed target cells to enable the enrichment of NeoPBL to frequencies comparable to cultured TIL over a 28-day period. Throughout this process, NeoPBL demonstrate specific reactivity against autologous tumor organoids and maintain memory-like features, including elevated expression of and . We additionally demonstrate that NeoPBL reactivity is polyclonal, encompassing multiple clonotypes that are detectable within TIL populations, underscoring physiological specificity for the targeted neoantigens. This streamlined process yields clinically relevant cell doses and enables identification and expansion of blood-derived neoantigen-specific TCRs. By potentially avoiding additional surgical risks and protracted delays of TIL and individualized TCR-engineered methods, the NeoPBL platform may have clinical and practical advantages. Ultimately, NeoPBL combines intrinsic cell fitness, minimal invasiveness and rapidity to potentially facilitate personalized adoptive cell therapy for cancer.

In the event of a large-scale radiological or nuclear emergency, a rapid, high-throughput screening tool will be essential for efficient triage of potentially exposed individuals, optimizing scarce medical resources and ensuring timely care. The objective of this work was to characterize the effects of age and sex on two intracellular lymphocyte protein biomarkers, BAX and p53, for early radiation exposure classification in the human population, using an imaging flow cytometry-based platform for rapid biomarker quantification in whole blood samples. Peripheral blood samples from male and female donors, across three adult age groups (young adult, middle-aged, senior) and a juvenile cohort, were X-irradiated (0-5 Gy), and biomarker expression was quantified at two- and three-days post-exposure.

Consistent sleep patterns are associated with better cardiovascular health, while sleep loss is known to impair vascular function. This study examined whether consistent sleep could improve vascular function and mitigate the negative effect of 25-hour total sleep deprivation. Sixteen healthy adults (10 females, 6 males; 34 ± 9 years; BMI: 25 ± 3 kg/m²) completed a randomized crossover study involving two 12-night sleep conditions, habitual sleep and a consistent sleep/wake schedule that were separated by a 1-2-week washout.

Bacoside A (BCA), a triterpenoid saponin isolated from Bacopa monnieri, exhibits diverse pharmacological properties, including neuroprotective, hepatoprotective, anti-stress, anti-inflammatory, and anti-ulcer effects. In the present study, BCA demonstrates pronounced anticancer activity against K562 chronic myelogenous leukemia (CML) cells by modulating autophagy-apoptosis dynamics. BCA induces dose- and time-dependent cytotoxicity in K562 cells while sparing normal human peripheral blood mononuclear cells (hPBMCs) and Vero cells, indicating therapeutic selectivity.
