A total of 93 adults with DS (aged 25-61 years) from the Alzheimer Biomarker Consortium-DS completed cognitive assessments, MRI and positron emission tomography (PET) scans (assessing amyloid-beta [Aβ] and tau), and a one-night home sleep study using the WatchPAT-300 device. Genome-wide association studies (GWAS) have enabled clinicians and researchers to identify genetic variants linked to complex traits and diseases(1). However, conducting GWAS remains technically challenging without bioinformatics expertise due to required data preprocessing, software installation, and analysis scripting (2,3). SAGA is a BASH-based, open-source, fully automated pipeline that integrates three widely adopted tools-PLINK(4), GMMAT(5), and SAIGE(6)-for accessible, robust, and reproducible GWAS.

Estimating heritability has been fundamental in understanding the genetic contributions to complex disorders like late-onset Alzheimer's disease (LOAD) and provides a rationale for identifying genetic factors associated with disease susceptibility. While numerous studies have established substantial genetic contribution for LOAD, the interpretation of heritability estimates remains challenging. These challenges are further complicated by the binary nature of LOAD status, where estimation and interpretation require additional considerations.

Occupational ApplicationsThe analysis of communication, attention, and cognitive engagement relative to leadership and non-technical skills (NTS) is important for understanding the team dynamic within clinical settings. We found negative correlations between passive communication (receiving information instead of dictating information) and leadership scores, suggesting that passive leadership traits lead to weaker technical leadership performance. Additionally, continuously sharing goals and providing affirmations to patients had positive relationships with leadership scores.
