To investigate whether metabotropic glutamate receptor 5 (mGluR5) in the basolateral amygdala (BLA) regulates inflammatory pain and pain aversion by mediating ERK/PI3K signaling pathway in mice. Two experimental routes were designed. In route 1, 18 male C57BL/6 mice were divided into control, IFA, and CFA groups. In route 2, 18 mice received stereotaxic BLA injections of ACSF, the mGluR5 agonist DHPG, or antagonist MTEP on day 8 following CFA administration. Pain behaviors and aversive emotions were assessed over 14 days. BLA tissues were collected on day 15 for immunofluorescence staining and Western blot analyses of mGluR5, PI3K, p-PI3K, and ERK1/2 expression. The expression levels of p-PI3K, mGluR5 and ERK1/2 in the BLA of CFA mice were significantly increased, suggesting that their activation was associated with inflammatory pain. DHPG increased the expression of p-PI3K and mGluR5 and ERK1/2 in the CFA mouse brain BLA, while MTEP had the opposite effect. Our results show that mGluR5 regulates inflammatory pain and pain aversion of CFA mice by mediating the ERK/PI3K signaling pathway.

This study aimed to identify the biomarkers that was associated with the postoperative incisional pain in patients with acute cholecystitis undergoing laparoscopic cholecystectomy surgery (ACC-LC). Sixty ACC-LC patients were enrolled and divided into mild pain (MP) and moderate-to-severe pain (MSP) groups based on their visual analog scale (VAS) scores 24 hours postoperatively. RNA sequencing was used to screen the potential pain associated markers, and ELISA were used to analyze the expression of one identified marker, CXCR5 in peripheral blood mononuclear cells (PBMCs).

Introduction: This study examined the impacts of different negative pressure cupping therapies (PCT) on pain relief, functional recovery, and inflammatory regulation in delayed onset muscle soreness (DOMS) after high-intensity exercise, with the aim of clarifying the dose-effect relationship.
Methods: In this study, 55 healthy male participants aged 18-25 were selected and divided into 5 groups: the control group (CTR; = 11) and NPCT groups at different levels (-25 kPa, -35 kPa, -45 kPa, and -55 kPa; = 11 in each group). A high-intensity protocol, which included 6 sets of lunges, squats, and squat jumps, was adopted to induce DOMS in the quadriceps femoris.
