For a long time neglected, the mpox virus has caused two public health emergency declarations by the World Health Organization in just two years. Rapid detection and characterization of the viral strains involved are crucial for better control of the disease. Recent work on viral genome sequencing in the Democratic Republic of the Congo (DRC) has shown the emergence in 2023 of clade Ib, with sustained human-to-human transmission and rapid spread to several other non-endemic neighboring countries, as well as outside Africa.

Mpox is a viral zoonotic disease characterized by flu-like symptoms followed by rash, which can be difficult to distinguish from other rash-presenting infections. Laboratory diagnosis is essential for confirming monkeypox virus (MPXV) infection; However, challenges in specimen transport and storage in endemic regions, especially in rural areas with limited infrastructure, hinder timely diagnosis. This study evaluates the use of SDS-EDTA-treated chromatography paper strips for testing different viral dilutions of MPXV, varicella-zoster virus (VZV), herpes simplex viruses 1 (HSV-1) and 2 (HSV-2) under different storage durations and temperatures.

Objectives: Understanding the antibody response in monkeypox virus (MPXV)-infected and uninfected individuals is essential for developing next-generation MPXV vaccines. This study aimed to characterize neutralizing antibody (NAb) and antibody-dependent cellular cytotoxicity (ADCC) responses in both groups, providing insights into immune protection and vaccine design.
Methods: A recombinant vaccinia Tian Tan (VTT) virus was utilized to develop high-throughput luciferase-reporter-based neutralization and ADCC assays.

Monkeypox virus (MPXV) is a poxvirus endemic to Central and West Africa with high epidemic potential. Poxviruses enter host cells via a conserved entry-fusion complex (EFC), which mediates viral fusion to the cell membrane. The EFC is a promising therapeutic target, but the absence of structural data has limited the development of fusion-inhibiting treatments.
