Article Abstract

This study investigates the use of machine learning (ML) techniques to predict intervention response in patients with Multiple Sclerosis (PwMS) using real-world data from wearable devices. Data from 27 PwMS, monitored over two months were analyzed with several state-of-the-art ML models to predict the efficacy of a computerized cognitive intervention targeting cognitive decline. A model based on Support Vector Machines achieved high accuracy in identifying patient response within the first 2-3 weeks of intervention, aided by feature selection methods like Mutual Information and Recursive Feature Elimination. Early prediction capability enables timely therapeutic adjustments, enhancing personalization of treatment and improving patient quality of life.

Initially developed for hematologic malignancies and multiple sclerosis, B-cell depletion therapies are now being investigated across a broader spectrum of autoimmune diseases, including glomerulopathies, both with and without associated podocytopathy. High morbidity and mortality associated with human β-coronavirus (CoV) infection highlight the need to determine host responses to infection and develop anti-viral therapies. Gap junction intercellular communication (GJIC), particularly involving Connexin43 (Cx43), is vital for maintaining central nervous system (CNS) homeostasis, and disruption of GJIC is a well-documented pathogenic mechanism among β-coronaviruses. Specifically, murine β-coronavirus, mouse hepatitis virus (MHV-A59) inoculation in the mouse brain causes acute-stage CNS viral spread and chronic neuroinflammatory demyelination while causing pronounced downregulation of Cx43 at the acute stage, reflecting a critical role in CNS pathology.
