Article Abstract

Chelation-assisted olefinic C-H functionalization has been demonstrated to be a powerful method of constructing multi-substituted alkenes from simpler ones. This strategy produces complex alkenes in a regio- and stereoselective manner, followed by C-H endo- and exo-cyclometallation. Among the various directing groups developed, N,N- and N,O-bidentate directing groups are the most widely used to selectively promote C-H functionalization due to their fine, tunable, and reversible coordination with the metal center. In this review, we discuss various N,N- and N,O-bidentate directing group-assisted olefinic C-H bond functionalization reactions, including alkenylation, alkylation, arylation, thiolation, silylation, halogenation, and cyclization.

The development of copper-catalyzed C-H functionalization processes is challenging due to the inefficiency of conventional chemical oxidants in regenerating the copper catalyst. This study details the development of a mediated electrosynthetic approach involving triple catalytic cycles in transient C-H functionalization to achieve efficient copper-catalyzed C-(sp)-H sulfonylation of benzylamines with sodium sulfinate salts. The triple catalytic system consists of a copper organometallic cycle for C-H functionalization, an aldehyde transient directing group (TDG) as an organocatalyst for imine formation, and a ferrocenium salt as an electrocatalyst.

We describe a photomediated protocol for the trifluoromethoxylation of benzylic, aldehydic, and non-activated C-H bonds, using bis(trifluoromethyl)peroxide (BTMP, (FCO)) as the key reagent. Under catalyst-free conditions in acetone, this reaction proceeds with selective functionalization of benzylic methylene groups. Furthermore, by using tetrabutylammonium decatungstate as a photocatalyst, the scope extends to include both non-activated methylene C(sp)-H and formyl C(sp)-H bonds.

An efficient synthesis and late-stage C-H functionalization of papaverine under volatile solvent-free conditions are reported. This methodology demonstrates significant potential for applications in the active pharmaceutical ingredient (API) industry, particularly for the sustainable and solvent-free synthesis of papaverine. A plausible reaction mechanism was meticulously elucidated through comprehensive control experiments.

A simple yet efficient method involving a visible-light-induced direct, regioselective chalcogenation of indoloquinoxaline derivatives has been developed. Thiols, disulfides and diselenides were found to be efficient as chalcogenating agents in the presence of Rose Bengal as a photosensitizer. This photoinduced C-H functionalization a cross-dehydrogenative-coupling (CDC) protocol was carried out at ambient temperature under an open-air atmosphere.

[2,1]-Azaboranaphthalenes represent unique boron-nitrogen (BN) isosteres of naphthalenes, attracting interest for the development of molecules with enhanced therapeutic potency. The existing synthetic strategies are generally two-component reactions with harsh conditions. Here we report an organocatalysed three-component modular synthesis of ring-fused BN isosteres and BN-2,1-azaboranaphthalenes following ring expansion of unstrained cyclic ketones (n = 4-8) via Wolff-type rearrangement.
