Characterization of lipid chain order and dynamics in asymmetric membranes by solid-state NMR spectroscopy.

Article Abstract

We studied the structure and dynamics of asymmetric POPC/(POPE/POPG) and POPS/(POPE/POPG) lipid membranes. To this end, the outer layer of multilamellar POPE/POPG (molar ratio 9 : 1) vesicles was exchanged (using methyl-β-cyclodextrin) by either chain deuterated POPC- or POPS-, for which H NMR order parameters were measured. As controls, we prepared symmetric POPC-/POPE/POPG and POPS-/POPE/POPG membranes of the composition of just the outer membrane of the asymmetric multilamellar vesicles and pure POPC- or POPS- multilamellar vesicles. Compared to symmetric membranes of the same lipid composition, chain order parameters () of the asymmetric preparations were higher in the upper half of the chain and lower in the lower half. This reshuffling of acyl chain order is also expressed in higher H NMR Zeeman order relaxation rates () of the chain segments in asymmetric membranes indicating alterations in the elastic properties of asymmetric bilayers as inferred from plots of . Asymmetric membranes showed increased stiffness and rigidity although the lipid acyl chain composition between the inner and outer leaflets were identical. There were no indications for chain interdigitation between the two leaflets in the NMR spectra, which led us to speculate that the interleaflet coupling could be accomplished by sensing the differences in lipid packing densities between the two leaflets. These alterations in leaflet properties should have consequences for lipid protein interaction and ultimately protein function.

Using the stable synthetic analogue 3-aza-dehydroxylysyl-phosphatidylglycerol (3adLPG), the putative role of native staphylococcal LPG in inhibiting the antibiotic daptomycin from binding to its target phosphatidylglycerol (PG), was investigated with respect to interfacial interactions between these lipids, daptomycin, and calcium ions. The influence of lipid monolayer/bilayer composition and interfacial ion concentrations upon the structure and integrity of model membranes were probed after daptomycin challenge using a combination of surface x-ray scattering techniques and fluorescence assays. In models representing the membrane composition of the daptomycin susceptible phenotype consisting of PG/3adLPG in a 7:3 M ratio, calcium ions drive the formation of two separate phases; Ca cross-linked PG/PG pairs and PG/3adLPG ion pairs.

Polyimide (PI) faces significant challenges in highly integrated and high-frequency electronic devices due to its inherently low thermal conductivity and relatively high dielectric constant (D). In this study, topologically micro-crosslinked PI films were synthesized by incorporating highly conjugated multi-amino polydiacetylene (MAPDA) into a fluorinated PI matrix. The unique alkene-alkyne alternating conjugated structure of MAPDA, combined with the strong electron-withdrawing trifluoromethyl groups in the matrix, promotes charge redistribution and reduces the dipole moment and polarizability.

Quasi-one-dimensional magnets can host an ordered longitudinal spin-density wave state (LSDW) in magnetic field at low temperature, when longitudinal correlations are strengthened by Ising anisotropies. In the S=1/2 Heisenberg antiferromagnet YbAlO_{3} this happens via Ising-like interchain interactions. Here, we report the first experimental observation of magnetization plateaux at 1/5 and 1/3 of the saturation value via thermal transport and magnetostriction measurements in YbAlO_{3}.
